摘要
目的:探讨PELP1与牙周膜细胞骨向分化的相关性。方法:通过对正常人牙周膜细胞与炎症人牙周膜细胞的分离、培养、鉴定,采用RT-PCR、免疫细胞化学、免疫荧光化学及Western blot技术,分别从基因和蛋白水平检测PELP1在两组细胞中的表达及其在细胞增殖和骨向分化过程中的表达变化。结果:在H-hPDLCs和I-hPDLCs中均存在PELP1 mRNA及蛋白的表达。PELP1参与调节hPDLCs的生长与增殖,在H-hPDLCs中与细胞增殖呈正相关,在I-hPDLCs中与细胞增殖呈负相关。PELP1的表达水平在两组hPDLCs骨向分化中亦表现出不同的变化趋势,在H-hPDLCs骨向诱导分化后升高,在I-hPDLCs骨向诱导分化后降低。结论:PELP1在炎症人牙周膜细胞中表达并参与调节细胞的生长增殖及骨向分化。
Objective:To investigate the relationship between PELP1 and bone differentiation in periodontal ligament cells. Methods:Human periodontal ligament cells( hPDLCs) were cultured and identified by immunocytochemistry. RT-PCR and Western blot were performed to detect PELP1 expression on proliferation and osteogenic differentiation. Results:The results showed that PELP1 was expressed in both H-hPDLFCs and Ⅰ-hPDLFCs. Compared with PELP1 expression in H-hPDLF-Cs,the PELP1 level of Ⅰ-hPDLFCs decreased with culture time. During osteogenic induction,the PELP1 expression in Ⅰ-hP-DLFCs was decreased with culture time,and similar with the expression of the osteogenic marker osteocalcin. Conclusion:PELP1 expressed in hPDLCs and played a role in PDLCs' proliferation and osteogenic differentiation.
