枸杞多糖诱导自噬抑制人舌鳞状癌细胞增殖 被引量:9

The autophagy and apoptosis induced by Lycium barbarum Polysaccharide in human tongue squamous cancer cells

摘要 目的:研究枸杞多糖体外抑制人舌鳞状癌CAL-27细胞增殖的效果及机制。方法:用不同浓度枸杞多糖分别作用CAL-27细胞,CCK-8法检测对细胞增殖的影响;流式细胞术检测细胞周期和凋亡变化;免疫荧光法检测自噬相关蛋白LC3的表达。结果:枸杞多糖呈剂量/时间依赖性抑制CAL-27细胞增殖,抑制率最高达25.19%;改变细胞生长周期,阻滞在G2期,并诱导细胞凋亡,呈剂量-反应关系。枸杞多糖调控LC3-Ⅰ型蛋白向LC3-Ⅱ型蛋白转化,诱导细胞发生自噬。结论:枸杞多糖可抑制人舌鳞状癌CAL-27细胞增殖,其机制可能与诱导细胞凋亡和自噬相关。 Objective:To investigate the effect and mechanism of Lycium barbarum Polysaccharides( LBP) on human tongue squamous cancer CAL-27 cells via induce autophagy.Methods:CAL-27 cells were treated with different concentration of LBP. Proliferation was tested by CCK-8. The apoptosis and cell cycle were analyzed by flow eytometry. Expression of autophagy-related protein LC3 was determined by immunofluorescence. Results:LBP could inhibit the proliferation of CAL-27 cells,induce cell apoptosis and arrest the cell cycle in the G2 phase with a dose-effect relationship. The LC3-Ⅱ protein expression following LBP treatments increased significantly with a dose-effect relationship,which suggested that LBP can regulate the expression of LC3 to induce autophagy. Conclusion:LBP might induce CAL-27 cell autophagy and apoptosis to significantly inhibit cell proliferation.
