热毒宁对脂多糖小鼠急性肺损伤和血红素加氧酶-1活性的影响

摘要 目的观察热毒宁注射液(reduning,RDN)对脂多糖(LPS)诱导小鼠的急性肺损伤(acute lung injury,ALI)保护作用,探讨其作用是否与血红素加氧酶-1(HO-1)有关。方法将48只雄性ICR小鼠平均随机分为Control组、RDN组、LPS组、LPS+地塞米松(DEX)组、LPS+RDN组、LPs+RDN+ZnPP(HO-1特异性抑制剂,锌原卟啉)组,每组8只。采用气管内滴注LPS(5mg/kg)诱导ALI;造模后24h,采集静脉血、取肺组织和支气管肺泡灌洗液(BALF),观察各组肺组织病理学改变,测量肺湿/干重比(W/D),BALF中的细胞总数和血清LDH活性、肺组织HO-1活性和HO-1蛋白表达量。结果与LPS组相比,LPS+RDN组大鼠肺组织损伤较轻,肺W/D、BALF中细胞总数和血清LDH活性显著降低,HO-1的活性和蛋白表达量显著升高,差异均有统计学意义(P〈0.05)。与LPS+RDN组相比,LPS+RDN+ZnPP组大鼠肺W/D、BALF中细胞总数、血清LDH活性显著升高,HO-1的活性和蛋白表达量减低,均差异有统计学意义(P〈0.05)。结论RDN对LPS诱导的ALI有保护作用,其保护作用与HO-1表达和活性增加有关。 Objective To investigate the effect of Reduning (RDN) injection on acute lung injury (ALI) and heme oxygenase-l(HO-1) in LPS-. induced mice. Methods 48 male ICR mice were randomly divided into six groups: control group, LPS group, RDN group, LPS+RDN group, LPS+DEX group and LPS+RDN+ZnPP group. Mice were intratracheal instillation with LPS (5mg/kg) to induce ALI. After LPS administration for 24 h, bronchoalveolar lavage fluid (BALF) and lung tissue were collected for the histopathological examinations. Lung wet/dry (W/D) ratio, total cell counts, the content of LDH, lung tissue HO-lactivity and HO-1 protein expression were examined. Results Compared with LPS group, lung W/D ratio, total cell counts and content of LDH were significantly decreased (P 〈0.05) while lung tissueHO-1 activity and HO-1 protein expression were significantly increased (P 〈0.05) in LPS+RDN group. The pathological changes of the lung were also significantly attenuated in LPS + RDN group and LPS + DEX group as compared to LPS group. ZnPP abolished the protective effects of RDN on LPS-induced ALL Conclusion RDN has protective effect on LPS-induced ALI, and its protective effect is related to the increase of HO-1 expression and activity.
