HIV感染无症状者外周血中HIV特异性T细胞及其亚群表达CD160情况的研究 被引量:1

Detection of HIV-specific T cells and their subsets expressing CD160 in peripheral blood of asymptomatic HIV-infected patients

摘要 目的 探讨HIV感染无症状者外周血中CD160在HIV特异性T细胞及其各亚群中的表达情况.方法 采集2014年6月至2015年11月于解放军第302医院就诊的43例未经抗病毒治疗的HIV感染无症状者和18例健康者的外周血,分离外周血单个核细胞,用免疫荧光染色法标记CD160后,以流式细胞仪检测其在HIV特异性T细胞及其各亚群(Tn、Tcm、Tem和Teff)细胞上的表达情况.结果 HIV感染无症状者外周血中,CD160在总体HIV特异性CD4 +T细胞和CD8+T细胞中表达的频数和MFI均较健康对照组高(P<0.01);HIV特异性CD4+T细胞的各亚群中,CD160在CD4+ Tn细胞中MFI,CD4+ Teff细胞中频数和MFI显著升高(P<0.01);HIV特异性CD8+T细胞的各亚群中,CD8+ Tn细胞、CD8 +Tcm细胞及CD8+ Tem细胞表达CD160的频数和MFI均显著升高(P<0.01).结论 慢性HIV感染可导致HIV特异性T细胞及其不同亚群中CD160高表达,引起细胞免疫功能紊乱,可能进而损伤了对HIV病毒的控制能力. Objective To investigate the expression of CD160 in HIV-specific T cells and their subsets in peripheral blood of asymptomatic human immunodeficiency virus (HIV) infected individuals.Methods A total of 43 asymptomatic HIV-infected individuals without antiretroviral therapy (ART) and 18 healthy controls were recruited from the No.302 Hospital of PLA between June 2014 and November 2015.Peripheral blood mononuclear cells (PBMC) were isolated from peripheral bloods of these subjects.CD160 was stained with fluorescent antibody.Expression of CD160 in HIV-specific T cells and their subsets was detected by flow cytometry.Results The frequency and median fluorescence intensity (MFI) of CD160 on the whole HIV-specific CD4 + T cells and CD8 + T cells were higher than those in the healthy controls (P〈0.01).In subsets ofHIV-specific CD4+ T cells,the MFI of CD160 in CD4+ Tn cells and CD4+ Teff cells,and the frequency of CD160 in CD4 + Teff cells were significantly increased (P 〈 0.01).CD8 + Tn fine,CD8 + Tcm cells and CD8 + Tem cells showed a significant upregulation in the frequency and MFI of CD160 (P 〈0.01).Conclusions Chronic HIV infection can lead to high expression of CD160 in HIV-specific T cells and their different subsets,causing cellular immune dysfunction,which may further impair the ability to control the HIV virus.
