生肌巴布贴对糖尿病皮肤溃疡小鼠创面愈合及愈合相关因子的影响

Impacts of Shengji Cataplasm on Wound Healing and Relevant Healing Factors in the Mice with Diabetic Skin Ulcer

目的探讨生肌巴布贴对糖尿病皮肤溃疡的修复作用。方法将40只小鼠全部建立糖尿病皮肤溃疡模型后,随机分为生肌巴布贴组、一效膏组、康复新液组、模型组四组,每组10只。生肌巴布贴组予生肌巴布贴(大小约2.0 cm×2.0 cm)贴于创面;一效膏组予一效膏均匀涂抹于创面,覆盖创面厚度约0.2 cm;康复新液组予康复新液均匀喷洒于创面;模型组以生理盐水涂抹于创面。各组于治疗后定期肉眼观察局部组织愈合情况,计算创面收缩率,并在治疗后第21天进行病理组织学检查,免疫组化检测血管内皮生长因子(VEGF)、纤维结合蛋白(FN)在局部组织中的表达情况。结果与模型组比较,生肌巴布贴组、一效膏组与康复新液组小鼠创面收缩率在用药后第5、11、19天均高于模型组,差异有统计学意义(P<0.05);生肌巴布贴组同一效膏组、康复新液组相比,差异无统计学意义(P>0.05)。治疗后第21天VEGF、FN表达情况与模型组相比,生肌巴布贴组、一效膏组和康复新液组在组织中单位面积阳性表达的平均光密度值较高(P<0.001)。生肌巴布贴组与一效膏组、康复新液组间比较差异无统计学意义(P>0.05)。结论新型制剂生肌巴布贴在促进创面愈合方面疗效与传统制剂一效膏相当。

Objective To discuss the repair effect of shengji cataplasm on diabetic skin ulcer.Methods A total of 40 mice were selected. After setting up the models of diabetic skin ulcer,all of them were randomized into a shengji cataplasm group,a yixiao ointment group,a kangfu liquid group and a model group,10 mice in each one. In the shengji cataplasm group,the shengji cataplasm（about 2. 0 cm × 2. 0 cm）was used on the wound site. In the yixiao ointment group,yixiao ointment was applied to the wound,about 0. 2 cm in thickness. In the kangfu liquid group,the kangfu liquid group was sprayed to the wound. In the model group,the physical saline was put on the wound. After treatment,the healing situation was observed visually at regular time. The wound healing rate was calculated. On the 21 stday after treatment,the histopathological examination was taken. The immunohistochemical test was adopted to determine the expressions of VEGF and FN in the local tissue. Results Compared with the model group,the wound healing rates were all higher the day 5,11 and 19 after treatment in the shengji cataplasm,yixiao ointment group and kangfuxin liquid group,indicating the significant differences（P〈0. 05）. The differences were not significant in the result of the shengji cataplasm group as compared with the yixiao ointment group and kangfuxin liquid group（P〉0. 05）.On day 21,in comparison of the expressions of VEGF and FN with the model group,the average optic density values of positive expression in the unit area were all higher in the shengji cataplasm,yixiao ointment group and kangfuxin liquid group,indicating the significant differences（P〈0. 05）. The differences were not significant in the result of the shengji cataplasm group as compared with the yixiao ointment group and kangfuxin liquid group（P〉0. 05）. Conclusion Shengji cataplasm has the similar therapeutic effects of wound healing as compared with the traditional yixiao ointment.
