大孔吸附树脂分离纯化2′-脱氧腺苷

摘要 主要研究了大孔吸附树脂分离纯化生物转化液中的2′-脱氧腺苷的工艺条件。通过静态吸附法考察了7种不同类型树脂的吸附能力,最适吸附树脂为D101,最佳吸附pH 8.0,确定了先用0.1mol/L Na2CO3洗脱再用体积分数30%乙醇洗脱的洗脱工艺。通过动态吸附实验确定了大孔吸附树脂分离纯化2′-脱氧腺苷的最佳工艺条件:上样体积流量2.0mL/min,洗脱体积流量1.5mL/min,洗脱剂依次为4.5倍柱体积的0.1 mol/L Na2CO3溶液、3倍柱体积的体积分数30%乙醇。在该条件下,质量分数78.7%的2′-脱氧腺苷收率为93.9%。 The technique of separation and purification of 2′-deoxyadenosine by macroporous adsorption resin was systematically investigated.The adsorption capacity of seven different kinds of resins were studied based on the static adsorption.The results showed that D101 resin was the optimal sorbent and the procedure of desorption were sequential filtration with 0.1 mol/L Na2 CO3 solution and 30%ethanol at pH 8.0.The optimum operating parameters were determined by dynamic adsorption as follows:sample flow rate of 2.0 mL/min,elution flow rate of 1.5 mL/min,eluting with 4.5 times column volume of 0.1 mol/L Na2 CO3,3 times column volume of 30%ethanol orderly.Under the condition,the content of 2-deoxyadenosine is as high as 78.7%,and the yield could reach to 93.9%.
