摘要 目的通过分析LGR5、SALL4、NKD1在乳腺癌组织与癌旁乳腺组织中的表达水平,探讨三者在乳腺癌组织中的表达及其相关性。方法选择2015年6月至2016年9月内蒙古包钢医院确诊并行乳腺癌切除术患者的乳腺癌组织30例及其对应的癌旁乳腺组织30例,年龄36~78岁,平均年龄57岁。应用Western blot从蛋白水平检测SALL4、LGR5、NKD1在乳腺癌组织及癌旁乳腺组织中的表达。结果 LGR5、SALL4、NKD1蛋白在乳腺癌组织表达分别为0.63±0.14、0.67±0.13、0.51±0.16(P<0.05),癌旁乳腺组织表达分别为0.53±0.16、0.58±0.11、0.61±0.11(P<0.05)。乳腺癌组织中LGR5与SALL4蛋白呈正相关(r=0.402,P<0.05);LGR5与NKD1蛋白呈负相关(r=-0.685,P<0.05);SALL4与NKD1蛋白呈负相关(r=-0.370,P<0.05)。LGR5、SALL4、NKD1蛋白在TNM分期中Ⅰ+Ⅱ期组表达分别为0.59±0.13、0.64±0.13、0.55±0.17(P<0.05),在Ⅲ期组中表达分别为0.75±0.10、0.75±0.09、0.41±0.11(P<0.05)。LGR5、SALL4、NKD1蛋白在淋巴结转移组中表达分别为0.73±0.11、0.74±0.08、0.40±0.11(P<0.05),在淋巴结未转移组中表达分别为0.58±0.13、0.64±0.14、0.57±0.16(P<0.05)。LGR5、SALL4、NKD1蛋白与乳腺癌患者的年龄、肿瘤的大小、肿瘤部位无关(P>0.05)。结论 LGR5、SALL4和NKD1的异常表达与乳腺癌发生、发展、浸润及转移密切相关,三者的联合检测可能对评估乳腺癌的生物学行为具有重要意义。LGR5、SALL4和NKD1可能是导致乳腺癌的发生、发展的重要因子。 Objective To evaluate the expression and the correlation of G-protein-coupled receptor 5(LGR5), sal-like 4(SALL4)and naked cuticle dro-sophila 1(NKD1) in breast cancer tissues and adjacent tissues, by analyzing their expression levels.Method From June 2015 to September 2016, 30 breast cancer patients with mastectomy, aged 36-78 years old with mean age of 57 years old were enrolled. Western blot was used to investigate the expression of LGR5, SALL4 and NKD1 in breast cancer tissues and corresponding adjacent breast tissues. Results The expressions of LGR5, SALL4 and NKD1 in breast cancer tissues were 0.63 ± 0.14, 0.67 ± 0.13 and 0.51 ± 0.16(P〈0.05), and in the adjacent tissues were 0.53 ± 0.16, 0.58 ± 0.11 and 0.61 ± 0.11(P〈0.05), respectively. Positive correlation was observed between LGR5 and SALL4 protein in breast cancer(r =0.402, P〈0.05). LGR5 was negatively correlated with NKD1 protein(r =-0.685, P〈0.05), and SALL4 was negatively correlated with NKD1 protein(r =-0.370, P〈0.05). The expression of LGR5, SALL4 and NKD1 protein in TNM staging Ⅰ + Ⅱwere 0.59 ± 0.13, 0.64 ± 0.13, 0.55 ± 0.17(P〈0.05), respectively, and expression in staging Ⅲ were 0.75 ± 0.10, 0.75 ±0.09, and 0.41 ± 0.11(P〈0.05), respectively. The expressions of LGR5, SALL4 and NKD1 were 0.73 ± 0.11, 0.74 ± 0.08 and0.40 ± 0.11(P〈0.05) in lymph node metastasis group, and were 0.58 ± 0.13, 0.64 ± 0.14, 0.57 ± 0.16(P〈0.05) in lymph node un-metastasis group. The expressions of LGR5, SALL4 and NKD1 protein were no associated with age, tumor size and tumor location in patients with breast cancer(P〈0.05). Conclusion It is demonstrated that abnormal expressions of LGR5, SALL4 and NKD1 are associated with the development and prognosis of breast cancer, and combined detection are important for as-sessing the biological characteristics of breast cancer. The regulation of LGR5, SALL4 and NKD1 in Wnt pathway may affect the development and progression of breast cancer.
