超高效液相色谱法测定大鼠血浆中α-萜品烯醇含量及其药代动力学研究

摘要 目的:用超高效液相色谱法(Ultra Performance Liquid Chromatography,UPLC)测定大鼠血浆中α-萜品烯醇含量,研究α-萜品烯醇在大鼠体内药物代谢情况。方法:SD大鼠灌胃给药,在给药后不同时间点取血,用UPLC检测不同时间点血样中α-萜品烯醇含量,绘制药时曲线,运用DAS.2软件计算药动学参数。结果:α-萜品烯醇的药代动力学最佳模型为一室模型,其主要药动学参数为AUC_((0→t))为(635.935±322.888)mg/(L·h),AUC_((0→∞))为(1 127.278±518.188)mg/(L·h),t_(1/2)z为(4.787±2.049)h,T_(max)为(0.5±0.329)h,C_(max)为(176.268±82.236)mg·L^(-1)。结论:该方法快速、简便、准确,可用于大鼠血浆中α-萜品烯醇的含量测定。 Objective: The aim of this study is to develop the quantitative method of α- terpineol in rat plasma by Ul- tra Performance Liquid Chromatography(UPLC) and investigate its pharmacokinetic for rats. Methods: The concentrations of paeonifiorin and albifiorin in plasma at different time after dosing were determined with UPLC method. The pharmaco- kinetic parameters were calculated by DAS. 2 pharmacokinetic program and the drug - time curve was drawn. Results : The optimal pharmacokinetic compartmental model of α- terpineol was one compartmental model. The pharmacokintic param- eters of α-terpineol were as follows : AUC(0→t) (635. 935 ± 22. 888) mg/( L h), AUC(0→∞) ( 1 127. 278 ±518. 188) mg/(L h), t1/2z (4.787±2.049) h, Tmax(0. 5 ±0.329) h, Cmax(176.268±82.236) mg L-1. Conclusion: It is proved that the method with UPLC is rapid, simple and accurate, which can be used for α-terpineol in rat plasma.
