摘要
目的:探讨痛泻要方缓解肠易激综合征腹泻(diarrhea-predominant irritable bowel syndrome,IBS-D)大鼠内脏高敏的机制。方法:将30只SD大鼠随机分为3组:空白组、IBS-D模型组及痛泻要方组。模型组和痛泻要方组通过结肠球囊扩张建立IBS-D内脏高敏模型。痛泻要方组按1 m L/100 g给大鼠灌胃痛泻要方溶液,1次/d,共28 d。结果:经痛泻要方治疗后,痛泻要方组大鼠的粪便含水量较模型组显著下降;内脏疼痛阈值则显著上升。痛泻要方组大鼠结肠黏膜内BDNFmRNA表达相比模型组显著下调,而在海马组织内则显著上调。结论:痛泻要方可能通过同时调控海马和结肠的BDNFmRNA表达来缓解IBS-D大鼠的内脏高敏现象。
Objective: To study the mechanism of Tongxie Yaofang regulating the expression of BDNFmRNA in diar- rhea- predominant irritable bowel syndrome (IBS- D) rats to ease visceral hypersensitivity. Method~: Thirty SD rats were randomly assigned to 3 groups : no intervention, IBS - D model or Tongxie Yaofang groups. IBS - D model was built with a balloon expansion in colon. The rats in the Tongxie Yaofang group were given solution of Tongxie Yaofang at a dose of 1 mL/100 g through gavage. The decoction was given once a day for 28 days. Results: Tongxie Yaofang significantly downregulated BDNFmRNA expression in colon tissue, while it significantly upregulated in hippocampus. Conclusion: Tongxie Yaofang may bi - directionally regulate both BDNFmRNA expression in colonic and hippocampus tissue to relieve visceral hypersensitivity in IBS -D rats.
