摘要
目的观察芪地消渴方对2型糖尿病气阴两虚证模型大鼠空腹血糖(fasting blood glucose,FBG)、C-肽(C peptide,C-p)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)及胰腺组织病理变化的影响,探讨芪地消渴方治疗2型糖尿病气阴两虚证的相关作用机制。方法将雄性SD大鼠77只随机分为7组,即正常组,模型组,芪地消渴方高、中、低剂量组,糖脉康组和优降糖组。用高脂饲料喂养,并以链脲佐菌素注射复制2型糖尿病模型,再用中药灌胃造成气阴两虚证。采用血糖测量仪检测给药前后FBG水平;采用放射免疫测定法检测血清中C-p、TNF-α、IL-6的水平,观察糖尿病模型大鼠胰腺组织病理形态学变化。结果与正常组比较,模型组大鼠FBG、TNF-α、IL-6水平显著升高(P<0.05),C-p水平显著降低(P<0.05)。与模型组比较,芪地消渴方高、中、低剂量组,消渴灵组和优降糖组大鼠FBG、TNF-α水平显著降低(P<0.05),C-p水平显著升高(P<0.05)。与模型组比较,芪地消渴方高、中剂量组,糖脉康组大鼠血清中IL-6水平显著降低(P<0.05);芪地消渴方低剂量组、优降糖组血清中IL-6水平与模型组比较,差异无统计学意义(P>0.05)。结论芪地消渴方可降低2型糖尿病气阴两虚证模型大鼠的FBG,升高血清中C-p水平,降低血清中TNF-α、IL-6水平,改善胰腺组织病理状态,改善胰岛素抵抗。
Objective To investigate the effect of Qidi Xiaoke prescription on fasting blood glucose (FBG), C-peptide (C-p), tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and histopathological changes of the spleen in a rat model of type 2 diabetes with deficiency of both qi and yin, as well as the mechanism of action of Qidi Xiaoke prescription in the treatment of type 2 diabetes with deficiency of both qi and yin. Methods A total of 77 male Sprague-Dawley rats were randomly divided into normal group, model group, high-, medium-, and low-dose Qidi Xiaoke prescription groups, Tangmaikang group, and glibenclamide group. High-fat feed and streptozotocin injection were given to establish a rat model of type 2 diabetes, and traditional Chinese medicine was given by gavage to induce deficiency of both qi and yin. A blood glucose meter was used to measure FBG before and after administration; radioimmunoassay was used to measure the serum levels of C-p, TNF-α, and IL-6; the histopathological changes of the spleen were ob- served in rats with diabetes. Results Compared with the normal group, the model group had significant increases in the serum levels of FBG, TNF-α and IL-6 (P〈0.05) and a significant reduction in the level of C-p (P〈0.05). Compared with the model group, the high-, medium-, and low-dose Qidi Xiaoke pre- scription groups, Tangmaikang group, and glibenclamide group had significant reductions in the levels of FBG and TNF-α (P〈0.05) and a significant increase in the level of C-p (P〈0.05). Compared with the model group, the high- and medium-dose Qidi Xiaoke prescription groups and the Tangmaikang group had a significant reduction, in the serum level of IL-6 (P〈0.05) ; there was no significant difference in the ser- um level of IL-6 .between the low-close Qidi Xiaoke prescription group/the glibenclamide group and the model group (P〈0.05). Conclusion In a rat model of type 2 diabetes with deficiency of both qi and yin, Qidi Xiaoke prescription can reduce FBG and the serum levels of TNF-a and IL-6, increase the serum level of C-p, improve the histopathological status of the spleen, and relieve insulin resistance.
