迷迭香酸乙酯对MPTP诱导的帕金森小鼠的保护作用 被引量:5

摘要 目的探讨迷迭香酸乙酯(Ethyl rosmarinate,ER)对MTPT诱导的帕金森小鼠的保护作用。方法用MPTP处理C57/BL6雄性小鼠用制作PD小鼠模型,通过行为学实验检测小鼠的自主活动、爬杆时间、游泳和悬挂能力。以Western blot的方法检测小鼠黑质TH蛋白的表达情况。以生化法测定小鼠中脑黑质取超氧化物歧化酶(SOD)、丙二醛(MDA)及谷胱甘肽-过氧化物酶(GSH-Px)得水平。结果 ER可以提高PD小鼠10min内的运行距离,缩短PD小鼠爬杆时间,增加PD小鼠游泳、悬挂能力评分,提高黑质TH蛋白的表达,降低i NOS的表达,提高脑组织SOD、GSH-Px酶活性,降低MDA含量。结论 ER对PD模型小鼠的治疗作用可能是通过抑制氧化损伤和炎症反应实现的。 Objective To study the neuroprotective effect of Ethyl rosmarinate( ER) against MPTP induced mouse model of Parkinson's disease. Methods Mice PD model was made by intraperitoneal administration of MPTP to C57/BL6 mice. Spontaneous movement,pole climbing time,swimming and traction ability were observed. TH protein expression was detected by Western blot. And the changes of SOD,GSH-Px activity and level of MDA simultaneously were detected. Results ER can significantly increase the traveling distance in 10 min,swimming and traction ability grade,decrease pole climbing times,increase the level of TH protein expression,increase the level of SOD and GSH-Px,decease the level of MDA. Conclusion ER can effectively inhibit the damage in DA neurons of PD mice,and the therapeutic action may related with its antioxidant and anti-inflammatory effects.
