摘要
目的应用高速离心分配色谱法(FCPC)分离纯化葛根中的大豆苷。方法以乙酸乙酯-正丁醇-乙醇-水(5:1:0.5:5)作为两相溶剂系统,上相为流动相,下相为固定相,转速1400 r/min,流速3.0 m L/min,检测波长254 nm,采用ascending模式进行分离制备,化合物的结构由质谱及核磁共振谱鉴定。结果从1.0 g葛根粗提物中分离得到16.3 mg大豆苷,纯度经HPLC分析为96.5%。结论该方法适合于葛根中的大豆苷的高效分离制备。
Objective To isolate and purify daidzin from the roots of Pueraria lobata by fast centrifugal partition chromatography( FCPC). Methods The experiment was performed with a two phase solvent system composed of ethyl acetate-n-butanol-ethanol-water( 5: 1: 0. 5: 5,v/v). The upper phase as mobile phase in ascending mode was operated at a rotation speed of 1400 r/min and the flow rate of 3. 0 m L/min with the detection wave length at 254 nm. The compound was analyzed by high performance liquid chromatography( HPLC),and elucidated by comprehensive NMR and ESI-MS analysis. Results A total 16. 3 mg of daidzin with the purity about 96. 5% was successfully obtained by FCPC from 1. 0 g of the crude extract of Pueraria lobata. Conclusion The FCPC was a simple,robust and efficient method for the preparation of daidzin from Pueraria lobata.
