摘要
荧光技术已发展成为分子生物学、细胞生物学、分析化学、环境检测和临床诊断的有力工具,具有灵敏度高,选择性好,时空分辨率高,设备简单廉价等突出优势。荧光探针通常由荧光基团和识别基团组成。识别基团可以选择性感知环境参数,也可以识别小分子、离子、酶、蛋白、核酸等。识别事件如何产生荧光响应,需要研究荧光发射的基本规律。当染料吸收光到达激发态后,其能量可以荧光形式释放,也可以被内转换、电子转移、能量转移、或系间穿越等过程耗散。识别事件通过对上述激发态能量释放途径的影响,从而产生荧光变化,实现对客体的荧光响应和检测。荧光探针的设计和使用涉及多学科,发展荧光探针对现代毕命科学和精准医学意义尤为重要。
