兔体内地高辛与胺碘酮相互作用

摘要 本文在健康家兔体内进行了地高辛与胺碘酮相互作用的研究,并比较血药浓度及动力学参数的变化。地高辛为灌服给药(115ng/kg),胺碘酮稀释后静脉注射(35mg/kg),采用拉丁方设计给药方案。结果:两药合用后地高辛的AUC增大,峰浓度升高,CL变化不明显;胺碘酮的AUC、Cmax、t_(1/2)的变化无显著意义,CL增高。 The interaction of digoxin-amiodarone wasstudied in 6 rabbits. Which were treated with latinsquare design in three groups and received a singledose of digoxin orally (115 ng/kg) or amiodaroneintravenously (35 mg/kg, diluted by 20 ml 5% dex-trose inj.) or the combination. The plasma concen-trations and portial pharmacokinetic parameters ofdigoxin and amiodarone were compared. The resultsindicate that by combination the area under theconcentration curve (AUC, ng·h/ml) and C_(max) ofdigoxin was increased, and CL showed no differ-ence. The CL of amiodarone was increased, butAUC (μg· h/ml), C_(max) and t_(1/2) showed no statisti-cally significant changes.
