HPLC法同时测定大鼠灌胃四物汤后血清阿魏酸和芍药苷含量

摘要 目的:建立HPLC同时测定大鼠灌胃四物汤后血清阿魏酸和芍药苷含量的测定方法。方法:24只大鼠分为空白对照组和四物汤低、高剂量组,各8只。空白对照组灌胃蒸馏水,四物汤低、高剂量组分别灌胃生药含量0.3 g/mL和0.6 g/mL的四物汤,灌胃体积为10 mL/kg,连续灌胃7 d。于最后一次灌胃1 h后水合氯醛麻醉,腹主动脉取血,制备含药血清,进行HPLC分析,紫外波长230 nm处检测。结果:阿魏酸和芍药苷的线性回归方程分别为Y=1.019 0X-0.031、Y=1.026X+0.091,r值分别为0.999 8、0.999 7,线性范围分别为0.62~18.03μg/mL、0.32~5.11μg/mL;精密度、专属性、稳定性和提取回收率考查均符合要求;四物汤高剂量组血清中的阿魏酸和芍药苷含量均明显高于四物汤低剂量组(P<0.01)。结论:该方法可以用于同时测定大鼠灌胃四物汤后血清中阿魏酸和芍药苷的含量。 ObjectiveTo establish the determination method of the serum contents of paeoniflorin and erulic acid in rats treated withintragastric administration of Siwu decoction using high performance liquid chromatography(HPLC). Methods:Twenty-four rats wererandomly divided into the control group,low and high dosage of Siwu decoction groups(8 rats each group). The control group,low andhigh dosage of Siwu decoction groups were treated with intragastric administration of distilled water,0. 3 g / mL and 0. 6 g / mL Siwudecoction(10 mL/ kg) for 7 days. The serum in three groups were harvested from abdominal aorta under chloralic hydras anesthesia,andanalyzed using HPLC at the 230 nm of wave length. Results:The linear regression equation,r value and linear range of paeoniflorin anderulic acid were Y = 1. 019 0X - 0. 031 & Y = 1. 026X + 0. 091,0. 999 8 & 0. 999 7 and 0. 62 to 18. 03 μg / mL & 0. 32 to 5. 11 μg /mL,respectively. The precision, specificity, stability and extraction recovery rate met with the requirement. The serum contents ofpaeoniflorin and erulic acid in high dosage of Siwu decoction group were significantly higher than those in low dosage of Siwu decoction(P 〈 0. 01). Conclusions:This method can determinate the serum levels of paeoniflorin and erulic acid in rats treated with intragastricadministration of Siwu decoction.
