摘要
为了探讨褪黑素对牛体外胚胎生产效果的影响,对体外成熟液(TCM199),受精液(BO)及胚胎培养液(SOF)添加不同浓度(0、10^(-8)、10^(-9)、10^(-10)褪黑素,研究在体外胚胎生产全程添加褪黑素对体外受精效果的影响。分别从屠宰场采集卵巢获得卵母细胞,卵母细胞经体外成熟、体外受精、早期胚胎体外培养。试验结果表明:体外胚胎生产过程中填加10^(-9)褪黑素组的卵母细胞卵裂率、囊胚率及囊胚细胞数显著优于其对照组(P<0.05);说明体外胚胎生产过程中全程填加抗氧化剂,可以达到抗细胞过氧化反应的目的,克服早期胚胎的发育阻滞,提高体外胚胎的囊胚发育率及囊胚细胞数,提高囊胚质量。
In order to investigate the effect of melatonin on cow embryo production in vitro effect of in vitro maturation,liquid(TCM199),by semen and embryo culture medium(BO)supplemented with different concentrations of melatonin(0,10^(-8),10^(-9),10^(-10),in vitro embryo production process to add the effect of melatonin on in vitro fertilization effect.The oocytes were collected from the slaughter house in cow ovary,oocyte in vitro maturation,in vitro fertilization and early embryo culture in vitro.The experimental results show that in the process of filling embryo production in vitro oocyte cleavage 10^(-9) melatonin group rate,blastocyst rate and blastocyst cell number was significantly better than the control group(P0.05);in vitro embryo production throughout the whole process of adding antioxidants.Can reach the purpose of anti peroxidation cell,overcoming early embryo developmental block,improve the in vitro embryo blastocyst rate and blastocyst cell number,improve the quality of blastocysts.
