摘要
利用Windows-BLAST软件同源比较分析获得地黄转录组中的DNA甲基转移酶/去甲基化酶基因,同时利用基因表达谱测序方法分析这些基因在地黄不同发育时期块根中的转录子水平。利用拟南芥11个C5-MTase和4个DMase基因的蛋白序列进行同源性比对,在地黄转录组中共获得了18个DNA甲基转移酶unigene和11个去甲基化酶unigene。差异表达基因(DEG)分析显示,11个DNA甲基转移酶unigene和5个去甲基化酶unigene在块根的发育过程中表达具有差异,随块根发育成熟转录子表达含量逐渐降低,说明甲基化基因参与调控地黄块根的发育。首次挖掘和分析了地黄基因组中DNA甲基转移酶/去甲基化酶基因的分布和表达,为进一步研究DNA甲基化在地黄块根发育以及连作下块根生长受抑的表观遗传学分子机理奠定了基础。
DNA methyltransferase and demethylation genes in the transcriptome of Rehmannia glutinosa were obtained by homol-ogy analysis using Windows-BLAST software. Meanwhile,the transcript levels of these genes in tuberous roots of different de-velopmental stages of Rehmannia glutinosa were analyzed by gene expression profiling. Using homologous sequences of11 C5-MTase and 4 DMase genes in Arabidopsis thaliana,18 DNA methyltransferase Unigenes and 11 demethylation enzyme Unigeneswere obtained in the transcriptome of Rehmannia glutinosa RNA-seq-based expression profiling showed that transcripts of 11 C5- MTase and 5 DMase unigenes decreased with the development of tuberous roots of R. glutinosa, indicating the involvement ofDNA methylation in root development of R. glutinosa. The distribution and expression of DNA methyltransferase and demethyl-ation genes m Rehmannia glutinosa genome were mvestigated and analyzed for the first dme,which research on the molecular mechanism of DNA methylation n the development of Rehmannia glutinosa root inhibition of root tuber under continuous cropping.
