摘要
目的探讨RhoE在人食管鳞状细胞癌中的表达及其与表皮生长因子受体(EGFR)表达的相关性,以及与患者的预后关系。方法选取2015年5月10日至2016年7月10日我院收治的食管鳞状细胞癌患者30例为研究对象,利用实时荧光定量聚合酶链反应(PCR)检测RhoE和EGFR基因在配对食管癌组织和正常食管组织中的表达;利用采集的患者生存时间,分析RhoE基因表达与患者预后的关系。结果与正常食管组织比较,食管癌组织中RhoE基因表达呈显著下降趋势,63.3%的组织呈RhoE表达下降大于2倍。EGFR表达在癌组织中上调,两者表达呈显著负相关(r=-0.550,P=0.032)。Kaplan-Meier生存分析表明,RhoE阴性表达患者生存期显著短于阳性表达者[风险比(HR)=0.210,P=0.038]。结论食管鳞状细胞癌中异常低表达的RhoE基因与EGFR表达负相关,且与患者更差的预后相关,提示RhoE基因在食管鳞状细胞癌中可能的抑癌作用。
ObjectiveTo investigate the gene expression of RhoE, a small Rho GTP protein in esophageal squamous cell carcinoma (ESCC) tissues and its association with epidermal growth factor receptor (EGFR) expression and patients prognosis.MethodsThirty patietns with confirmed diagnosis of ESCC were included in this study form May 10 2015 to July 10 2016. Real time quantitative PCR was applied to detect the expression of RhoE and EGFR in paired ESCC and corresponding normal tissues. The prognosis of RhoE was further evaluated based on the recorded survival time of the 30 ESCC patients.ResultsComparing with the normal esophageal tissues, RhoE was significantly downregulated in ESCC. 63.3% of these ESCC tissues expressed more than 2 fold downregulation of RhoE. Besides, EGFR gene was upregulated in these corresponding ESCC tissues. A significant negative correlation was found between RhoE and EGFR expression (r=-0.550, P=0.032). Kaplan-Meier analysis of association between RhoE expression and overall survival of these ESCC patients revealed that lower RhoE levels indicated poorer prognosis [hazard ratio (HR)=0.210, P=0.038].ConclusionDown-regulation of RhoE was observed in ESCC tissues, which negatively correlated with EGFR expression and prognosis. All these indicate the vital tumor suppressing roles of RhoE in the development of ESCC.
