摘要 目的:应用Box-Behnken效应面对重楼中重楼总皂苷的提取工艺进行优化研究。方法:将提取工艺中乙醇的浓度、重楼总皂苷提取时间及液药比作为自变量,重楼总皂苷提取率为因变量,用效应面法选择最佳提取工艺,并将预测值与观测值进行比较。结果:重楼中重楼总皂苷的最佳提取工艺参数采用62.5%浓度乙醇、液药比例为16.5∶1,提取时间为3.0h。经3批工艺验证验证重楼总皂苷的实际提取量与预测提取量二者的相对误差为2.61%。结论:使用Box-Behnken效应面法优化重楼中重楼总皂苷的提取工艺,方法简便,结果可靠,预测性良好。 Objective:To optimize the extraction process of total saponins by Box-Behnken effect surface.Methods:The extraction rate of total saponins was determined by the regression method of ethanol concentration,extraction time and ratio of material to liquid,and the extraction rate of the tota saponin.And compared the predicted value with the observed value.Results:The optimum extraction parameters were as follows:the concentration of ethanol was 62.5%,the ratio of liquid to material was 16.5∶1,the extraction time was 3.0 h,the extraction parameters were obtained by three batches,and the extraction amount of total saponins The relative error between the predicted value and the regression model is 2.61%.Conclusions:The Box-Behnken effect surface method is used to optimize the extraction process of the total saponins in the re-building.The method is simple,the result is reliable and the prediction is good.
