摘要
目的探讨昆仑雪菊水提物对饱和脂肪酸棕榈酸(PA)诱导胰岛β细胞损伤的保护作用。方法以大鼠胰岛β细胞系INS-1细胞为实验细胞,采用棕榈酸诱导建立INS-1胰岛β细胞损伤模型,给予不同浓度(0.1、1、5、10μg/mL)的昆仑雪菊水提物进行干预,利用克隆形成实验、CCK8法等观察昆仑雪菊水提物对高脂作用下胰岛β细胞凋亡的影响,并运用DAPI染色及AnnexinV/PI双染测验INS-1细胞的凋亡情况。结果昆仑雪菊水提物对正常INS-1细胞生长具有双向调节作用,低浓度(0.1~10μg/mL)促进细胞的增殖,高浓度(50~250μg/mL)抑制细胞增殖;棕榈酸呈剂量依赖性地诱导INS-1细胞凋亡,昆仑雪菊水提物(1、5、10μg/mL)能明显拮抗棕榈酸诱导的INS-1细胞凋亡。结论昆仑雪菊水提物在一定剂量范围内(1、5、10μg/mL)能促进胰岛β细胞增殖,对棕榈酸诱导的胰岛β细胞凋亡具有保护效应。
Objective To investigate the protective effects of aqueous extract of Xinjiang Coreopsis tinctoria flowers(XCTF)on pancreatic Isletβcells injury induced by Palmitic Acid(PA).Methods Pancreaticβcell line INS-1 were given long-term induction with PA to establish the impaired cell model,and then were intervened with aqueous extract of XCTF.Cell viability was determined by tetrazolium salt(XTT)colorimetry.Cell apoptosis ratio were detected by flow cytometry.Results The aqueous extract of XCTF had the bidirectional regulation effect on the growth of normal INS-1 cells,and the low concentration promoted cell proliferation and high concentration inhibits cell proliferation;PA impaired the viability of INS-1 cells in concentration-dependent manner,and the aqueous extract of XCTF improved the impairment induced by PA in concentration-dependent manner.Moreover,the aqueous extract of XCTF improved the injury well especially on serious injury in the INS-1 cell by PA.Conclusion The aqueous extract of Xinjiang Coreopsis tinctoria flowers can promote pancreatic isletβ-cell proliferation within the certain dose range,and has a protective effect on pancreatic isletβ-cell injury induced by PA.
