RAGE在人胃癌组织的表达及其与临床病理特征的关系 被引量:1

The expression of RAGE in human gastric cancer tissue and its relationship with clinicopathological characteristics

摘要 目的探讨晚期糖基化终末产物受体(receptor for advanced glycation end products,RAGE)在胃癌组织、癌旁组织、正常胃组织中的表达差异及RAGE表达与胃癌临床病理学特征的关系。方法收集胃癌组织和癌旁组织各180例,及正常胃组织30例,应用免疫组织化学(immunohistochemistry,IHC)染色方法检测RAGE表达情况,并分析RAGE与胃癌临床病理学特征的关系。结果胃癌组织、癌旁组织正常胃组织中RAGE表达阳性率分别为70.0%(126/180)、45.0%(81/180)、40%(12/30),胃癌组织中RAGE表达比癌旁组织及正常胃组织中的表达显著增强(P<0.05),而癌旁组织及正常胃组织中RAGE表达差异无统计学意义(P>0.05)。RAGE表达在年龄、性别、肿瘤的原发部位方面差异无统计学意义(P>0.05);随着高、中、低分化程度的降低,RAGE阳性表达率逐渐升高,差异有统计学意义(P<0.05);按胃癌TNM分期,随着T1至T4期浸润深度增加,其阳性表达率逐渐增高,差异有统计学意义(P<0.05);有淋巴结转移胃癌组织中RAGE阳性表达率(78.0%,103/132)明显高于无淋巴结转移胃癌组织中RAGE阳性表达率(47.9%,23/48),差异有统计学意义(P<0.05);无远处转移143例,其RAGE阳性表达率为65.7%(94/143),有远处转移37例,其RAGE阳性表达率为86.5%(32/37),有远处转移者RAGE阳性表达率(86.5%,32/37)高于无远处转移者RAGE阳性表达率(65.7%,94/143),二者差异有统计学意义(P<0.05)。结论 RAGE在胃癌组织中高表达,而在癌旁组织和正常胃组织中低表达;RAGE的高表达与胃癌的分化程度、侵润深度、是否有淋巴结转移、是否有远处转移相关,是胃癌治疗的潜在靶点。 Objective To study the expression level and significance of receptor for advanced glycation end produ(RAGE)in gastric cancer tissues,adjacent tissues and normal gastric tissues,and to explore the relationship between the RAGE protein levels and the characters of the patients.Methods Immunohistochemical(IHC)staining method was used to detect the expression of RAGE of 180 cases of gastric cancer,180 cases of adjacent tissue and 30 cases of normal gastric tissues and analyze the relationship between RAGE and clinical pathological features of gastric cancer.Results Positive rates of RAGE expression of the gastric cancer tissue,the adjacent tissue and the normal tissue were 70.0%(126/180),45.0%(81/180),40%(12/30),RAGE expression in the gastric cancer tissue was higher than in the adjacent tissue and the normal gastric tissue(P <0.05),and the difference of RAGE expression in the adjacent tissues and normal gastric tissue had no statistical significance.There was no statistical difference in age,gender,and tumor origin.The positive expression rate of RAGE increased gradually with the decrease of differentiation degree,and the difference was statistically significant.According to the TNM stage of gastric cancer,the positive expression rate increased gradually with the increase of the infiltration depth of T1 to T4,and the difference was statistically significant.Tthe RAGE positive expression rate(78.0%,103/132)in gastric cancer tissue with lymph node metastasis were significantly higher than those(47.9%,23/48)without lymph node metastasis,and the difference was statistically significant(P <0.05);there were 143 cases without distant metastases,the RAGE positive expression rate was 65.7%(94/143),whereas 37 cases with distant metastasis,the RAGE positive expression rate was 86.5%(32/37),and the difference was statistically significant(P <0.05).Conclusion The RAGE protein expression was higher in gastric cancer tissues than that in adjacent tissues and normal gastric tissues;the high expression of RAGE protein is related to the degree of malignancy of gastric cancer,such as differentiation degree,invasion depth,lymph node metastasis,and whether there is lymph node metastasis and distant metastasis,which will be a potential target for gastric cancer treatment.
