摘要
目的测定神香草30%和70%乙醇提物中迷迭香酸和绿原酸含量。方法采用回流提取法,分别制备神香草30%和70%乙醇提物,采用高效液相色谱法,色谱条件:Sapphire C18色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.2%醋酸(48∶52),检测波长330nm,柱温30℃,流速1.0mL/min,进样量10μL,进行梯度洗脱分离检测。结果在该色谱条件下神香草乙醇提取物中迷迭香酸、绿原酸分离良好,其线性范围分别为12.96~95.20μg/mL(r=0.999 6)、10.89~75.35μg/mL(r=0.999 7),回收率分别为99.56%、101.4%,RSD分别为1.3%、1.2%。70%乙醇提取物中迷迭香酸和绿原酸的含量分别为6.404、2.286mg/g,30%乙醇提取物中迷迭香酸的含量为4.81mg/g,绿原酸含量很少(不在绿原酸对照品线性范围之内)。结论该方法分离效果好、重复性好、准确,适用于神香草中迷迭香酸、绿原酸的含量测定。
Objective To determine contents of rosmarinic acid and chlorogenic acid from 30% and 70%ethanol extracts of Hyssopus officinalis L(HOL).Methods Different volume fractions of ethanol were used in hot reflux extraction to extract the HOL,and rosmarinic acid and chlorogenic acid was analyzed by HPLC,applying Sapphire C18 column with acetonitrile-0.2% acetic acid solution as mobile phase and10μL of sample size running at 1.0 mL/min under 330 nm at 30℃ column temperature via gradient eluting.Results The contents of rosmarinic acid and chlorogenic acid from 70% ethanol extracts of HOL were 6.404 mg/g and 2.286 mg/g respectively.The contents of rosmarinic acid from 30% ethanol extracts of HOL were 4.81 mg/g,but chlorogenic acid content was very little(not within the linear range of chlorogenic acid reference substance).Conclusion The method has good separating efficacy,repeatable and accurate,which is applicable to determining contents of rosmarinic acid and chlorogenic acid from different ethanol extract of Hyssopus officinalis L.
