不同质量浓度医用臭氧对兔骨骼肌损伤组织学与IL-6的影响 被引量:7

摘要 目的研究不同质量浓度的医用臭氧(O3)局部注射对兔骨骼肌损伤组织的修复效果及组织与血浆IL-6水平的影响,评估不同质量浓度医用臭氧对骨骼肌急性损伤的疗效与毒性。方法 90只实验兔随机分为5组:分为空白对照组、模型组、臭氧治疗Ⅰ组(20μg/ml)、Ⅱ组(30μg/ml)、Ⅲ组(50μg/ml)。模型组和臭氧组均采用骨骼肌挤压伤造模器制备骨骼肌挤压伤模型,臭氧治疗组在创伤局部注射不同浓度臭氧治疗。通过HE染色观察治疗后24 h、5 d、10 d骨骼肌组织学改变和组织与血浆IL-6水平。结果组织学改变:在24 h模型组、治疗Ⅰ组有大面积横纹肌肿胀及肌纤维断裂溶解,治疗Ⅱ、Ⅲ组中横纹肌病变的程度及范围均有减轻;5 d时各组中病变有不同程度的减轻,尤其是Ⅱ、Ⅲ组中受损的横纹肌细胞形态大致正常;10 d时模型组及治疗Ⅰ组损伤以慢性炎症反应为主,且Ⅰ组炎症反应相对较轻;Ⅱ、Ⅲ组则以横纹肌细胞退行性变为主,Ⅲ组中见弥漫横纹肌细胞退行性变及部分肌细胞萎缩;IL-6表达量:模型组与Ⅰ组在24 h、5 d和10 d均显著高于空白组,Ⅱ和Ⅲ组在24 h和5 d均显著高于空白组;臭氧治疗组在24 h、5 d和10 d均显著低于模型组;Ⅲ组在24 h和5 d均显著低于Ⅰ和Ⅱ组,10 d显著低于Ⅰ组,与Ⅱ组比较无统计学差异;Ⅱ组在24 h、5 d和10 d均显著低于Ⅰ组。结论 20μg/ml、30μg/ml和50μg/ml的臭氧均可修复损伤的骨骼肌,50μg/ml的臭氧有肌细胞损伤,以30μg/ml对骨骼肌损伤的修复效果最佳。 Objective To evaluate the effect and toxicity of medical ozone onto skeletal muscle damage and level of IL-6 by local injections with ozone at different concentration. Methods 90 rabbits were randomly divided into blank control group,model group,ozone treatment groupⅠ( 20 μg/ml),group Ⅱ( 30 μg/ml) and group Ⅲ( 50 μg/ml). In the model group and the ozone group were preparated skeletal muscle crush injury model. In the ozone treatment group were given different concentrations of ozone therapy. HE staining was used to observe the histological changes of skeletal muscle and enzyme linked immunosorbent assay to detect the content of IL-6 in plasma. Results The histological changes: in the 24 h model group,a large area of muscle swelling and muscle fiber fracture dissolution were found in the treatment group I,the range and degree of treatment Ⅱ and group Ⅲ in striated muscle lesions were lessened; in 5 d,lesions in each group had different degrees of ease,especially in group Ⅱ and Ⅲ,damaged striated muscle cells were generally normal; in 10 d,main injury of model group and treatment group I was chronic inflammation,and the inflammatory reaction in group I was relatively light; while in group Ⅱ and Ⅲ,muscle cell degeneration could be seen,and in group Ⅲ,diffuse degeneration of striated muscle cells and muscle cell atrophy were found. The expression of IL-6 : the expression of IL-6 in model group and group I in 24 h,5 d and 10 d were significantly higher than that of the control group,those of group Ⅱ and Ⅲ in 24 h and 5 d were significantly higher than that of the control group. Ozone treatment group in 24 h,5 d and 10 d were significantly lower than that of the model group; the expression of IL-6 in group Ⅲ in 24 h and 5 d were significantly lower than those of group I and Ⅱ. 10 d was significantly lower than that in group I,had no significant difference with group Ⅱ; Ⅱ group in 24 h,5 d and 10 d were significantly lower than those in group I. Conclusion 20 μg/ml,30 μg/ml and 50 μg/ml of ozone can repair the damage of skeletal muscle,50 μg/ml of ozone is with muscle cell damage,30 μg/ml of ozone on skeletal muscle injury repair effect is the best.
