摘要
目的建立四味黄连洗剂的UPLC指纹图谱及指标性成分的含量测定方法。方法采用Acquity UPLC HSS T3色谱柱(50 mm×2.1 mm,1.8μm),柱温30℃,流速0.25 mL·min^(-1),检测波长242 nm,流动相为乙腈(A)-0.1%磷酸水溶液(B),梯度洗脱;利用《中药色谱指纹图谱相似度评价系统(2004A版)软件》,比较分析了10批四味黄连洗剂的色谱图,同时测定了四味黄连洗剂中黄芩苷、盐酸小檗碱和大黄素的含量。结果建立了四味黄连洗剂的特征指纹图谱,并利用对照品指认了13个共有峰中的3个主要共有峰。黄芩苷、盐酸小檗碱、大黄素的线性范围分别为72~600 ng(r=0.9994)、72~600 ng(r=0.9997)、1.2~10 ng(r=0.9994);平均回收率分别为92.95%、99.37%、93.34%,RSD分别为1.16%、1.46%、0.88%。结论所用方法具有良好的精密度、稳定性和重复性,可用于四味黄连洗剂的质量控制。
OBJECTIVE To establish an UPLC fingerprint and a method for the determination of index components of Siwei Huanglian lotion( SWHLL). METHODS The UPLC fingerprint of SWHLL were determined on Acquity UPLC HSS T3 (50 mm×2.1 mm, 1.8 μm) , eluted with mobile phase containing of aeetonitrile - 0. 1% phosphoric acid in a gradient mode. The temperature of column was 30℃,the flow rate was 0.25 mL.min-1 and the detection wavelength was 242 nm. The chromatograms of 10 batches of SWHLL were compared using the software Similarity Evaluation System for Chromatographic Fingerprint of traditional Chinese medicine (version 2004A). The contents of baicalin,berberine hydrochloridein and emodin were also determined simuhaneously. RESULTS The UPLC fingerprint of SWHLL had been established. There were thirteen common peaks. Three chromatographic peaks in the common peaks were identified by reference substances. The linear ranges of baicalin, berberine hydrochlofide and emodin were 72 -600 ng( r = 0. 9994) ,72- 600 ng (r = 0. 9997 ) and 1.2 - 10 ng (r = 0. 9994). The average recovery was 92.95% , 99.37% and 93.34% respectively,and the RSD was 1. 16% , I. 46% and 0.88% respectively. CONCLUSION This method has desirable precision, stability, repeatability, and can be applied to quality control of SWHLL.
