山葡萄酒MLF乳杆菌最佳培养基的筛选

Screening of the Best Medium of MLF Lactobacillus

以OD_(600)值作为评价指标,对MRS和改良M17培养基进行比较选择出基础培养基,在基础培养基中添加不同的果汁探究不同果汁对MLF乳杆菌的影响;采用单因素和正交试验对果汁加量、碳源、总氮源和盐液A进行优化,得出最佳组合为:大豆蛋白胨为5.4g/L,胰蛋白胨为5.4g/L,牛肉蛋白胨为3.6g/L,酵母浸粉为3.6g/L,K_2HPO_4为0.7g/L,抗坏血酸为0.5g/L,盐液A为2g/L,甘油为8g/L,乳糖为8g/L,葡萄汁为130g/L。此培养基培养MLF乳杆菌,其培养液的OD_(600)值比改良M17培养基高大约32.2%。

The test used OD600 value as the evaluation index, we elected better medium with comparingthe modified M17 medium to MRS medium. Adding different juice is to explore the effect of different juice onlactobacillus. By single factor and orthogonal, it could be optimized that involved the content of juice, C source,N source and A salt solution. It could be the best optimum combination of culture medium ： soya peptone 5.4g/L, tryptone 5.4g/L, beefpeptone 3.6g/L, yeast extract powder 3.6g/L, potassium hydrogen phosphate 0.7g/L, L- ascorbic acid 0.05g/L, the salt solution A 2g/L, glycerol 8g_/L, lactose 8g/L, grape juice 130g/L. Thismedium cultivates MLF lactobacillus, OD600 values of the culture medium about 32.2 % is higher than the im-proved M17 medium.
