宫颈组织中白介素-2、白介素-4在HPV感染不同类型宫颈病变中的表达

Expression and Significance of IL-2,IL-4 in Different Types of Cervical Lesions With HPV Infection

目的探究与分析IL-2、IL-4在HPV感染不同程度宫颈病变中的表达及意义。方法选取我院自2015年3月—2017年3月收治的40例行宫颈活检组织切片显示为正常宫颈组织作为正常组,另选择同时期收治的行宫颈活检组织切片显示的CINⅠ12例,CINⅡ10例,CINⅢ9例,宫颈浸润性鳞癌组织30例。采用原位杂交法检测上述样本中IL-2、IL-4及HPV16/18的表达水平。结果 IL-2在HPV16/18阳性表达的正常组、CIN组及鳞癌组中阳性表达率相比,差异具有统计学意义(P<0.05)。即IL-2的表达与宫颈病变程度呈负相关(r=-0.783,P<0.05)。IL-2在HPV16/18阴性表达的正常组、CIN组及鳞癌组中阳性表达率相比,差异无统计学意义(P>0.05)。IL-4在HPV16/18阳性表达的正常组、CIN组及鳞癌组中阳性表达率相比,差异具有统计学意义(P<0.05)。即IL-4的表达与宫颈病变程度呈正相关(r=0.668,P<0.05)。IL-4在HPV16/18阴性表达的正常组、CIN组及鳞癌组中阳性表达率相比,差异无统计学意义(P>0.05)。结论在HPV感染宫颈组织后可见局部IL-2表达水平降低,IL-4表达水平升高,提示发生HPV感染宫颈的原因可能与宫颈局部免疫细胞水平降低有关。

Objective To explore and analyze the expression and significance of IL-2, IL-4 in different types of cervical lesions with HPV infection. Methods 40 routine cervical biopsys from March 2015 to March 2017 were displayed by our hospital diagnosed normal tissue as the normal group. At the same time, 12 cases of chronic cervicitis were displayed by our hospital diagnosed the CIN Ⅰ, 10 cases were diagnosed CIN Ⅱ, 9 cases were diagnosed CIN Ⅲ, 30 cases of cervical invasion of squamous carcinoma were diagnosed. the level of the IL-2, IL-4 and the expression of HPV16/18 were detected by in situ hybridization method. Results The positive expression rate of IL-2 in HPV16/18 positive expression group, CIN group and squamous cell carcinoma group were compared, the difference was statistically significant（P〈0.05）. There was a negative correlation between the expression of IL-2 and the degree of cervical lesions（r=-0.783, P〈0.05）. There was no significant difference in the positive expression rate of IL-2 between HPV16/18 negative expression group, CIN group and squamous cell carcinoma group（P〈0.05）. The positive expression rate of IL-4 in HPV16/18 positive expression group, CIN group and squamous cell carcinoma group were compared, the difference was statistically significant（P〈0.05）. There was a positive correlation between the expression of IL-4 and the degree of cervical lesions（r=0.668, P〈0.05）. There was no significant difference in the positive expression rate of IL-4 between HPV16/18 negative expression group, CIN group and squamous cell carcinoma group（P〈0.05）. Conclusion In the cervical organization with HPV infection, the level of IL-2 expression has decreased, However the level of IL-4 has risen, which may reveal that cervical HPV infection in the tip may be associated with the drop of cervical local immune cell levels.
