摘要
目的观察瑞香素对2型糖尿病(T2DM)大鼠血清炎症因子水平的影响以及对血糖、血清胰岛素的作用,探讨瑞香素对改善糖尿病大鼠胰岛素抵抗的作用机理。方法予健康雄性Wistar大鼠高糖高脂饲料喂养30天后,腹腔注射链脲佐菌素(STZ)40mg/kg制备T2DM大鼠模型,将造模成功大鼠随机分为模型组(12例)和瑞香素组(12例),同时将正常大鼠随机分为正常组(12例)和对照组(12例)。分别予对照组和瑞香素组大鼠腹腔注射瑞香素每天4mg/kg,共注射30天,检测各组大鼠血清炎症因子、空腹血糖(FPG)和空腹胰岛素(FINS)水平,并计算胰岛素抵抗指数(HOMA-IR)。结果与正常组相比,模型组大鼠血清白细胞介素(IL)-6和肿瘤坏死因子(TNF)-α均明显升高(P〈0.01),FPG、FINS水平以及HOMA—IR也明显升高(P〈0.01)。瑞香素组大鼠的各项指标较模型组均明显降低(P〈0.05)。正常组与对照组上述指标比较差异无统计学意义(P〉0.05)。结论瑞香素可能通过降低T2DM大鼠血清炎症因子水平从而起到降低血糖和改善胰岛素抵抗的作用。
Objective To observe the effects of daphnetin on the serum levels of serum imflam- matory cytokines ,blood glucose and insulin in rats with type 2 diabetes mellitus (T2DM) and to explore the mechanism for its improvement on insulin resistance of rats. Methods T2DM rat model was induced by intravenous injection of streptozotocin(40 mg/kg) after 30 days with high glucose and high fat diet. T2DM model rats were randomized into the model group (n = 12 ) and the daphnetin group (n = 12 ). Meanwhile,normal rats were assigned to the normal group( n = 12) and the control group( n = 12). The daphnetin group and the control group were injected intraperitoneally with daphnetiu 4 mg/kg once a day for 30 days. And then serum imflammatory cytokines, fasting plasma glucose (FPG), fasting insulin (FINS) and insulin resistance index ( HOMA-IR ) of all the rats were measured. Results Compared with the normal group, interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) of the model group were significantly bigher(P 〈 0.01 ) as well as FPG, FINS and HOMA-IR (P 〈 0.01 ). Compared with the model group, above indexes were significantly lower in the daphnetin group ( P 〈 0.05 ). No significant differences of above indexes between the normal group and the control group were found ( P 〉 0.05 ). Conclusion Daphnetin can lower blood glucose and reduce insulin resistance by decreasing the level of inflammatory cytokines in T2DM rats.
