瘢痕灸肝俞穴、足三里穴对原发性肝癌大鼠Wnt10b、Wnt3a表达的影响

Effects of Scarring Moxibustion Ganshu-point and Zusanli-point on Expression of Wnt10B and Wnt3A of Hepatic Carcinoma Rats

目的:探讨瘢痕灸肝俞穴、足三里穴对原发性肝癌大鼠Wnt/β-catenin信号传导通路中Wnt3a、Wnt10b表达的影响。方法:健康雄性Wistar大鼠随机分成对照组、模型组、阿霉素组、肝俞组、足三里组,通过注射二乙基亚硝胺制备肝癌模型,用RT-PCR检测各组Wnt3a、Wnt10b表达量,观察肝细胞结构和形态变化。结果:与对照组比较,模型组、阿霉素组、肝俞组和足三里组Wnt3a、Wnt10b表达量均显著升高;与模型组比较,阿霉素组、肝俞组和足三里组表达量降低;与阿霉素组比较,肝俞组、足三里组表达幅度下降;与肝俞组比较,足三里组表达量升高。镜下观察可见,对照组为正常肝细胞,模型组细胞有假小叶形成,阿霉素组有明显增生灶,肝俞组细胞核多形、核浆部分消失,足三里组纤维样变。结论:瘢痕灸肝俞穴、足三里穴对原发性肝癌大鼠有治疗效果,其机制可能是通过调节Wnt/β-catenin信号传导通路中Wnt3a、Wnt10b的表达量来实现的。

Objective： To explore the scarring moxibustion Ganshu-point（BL18）,Zusanli-point（ST36） on expression of Wnt3a,Wnt10b of Wnt/β-catenin signaling pathway of hepatic carcinoma rats.Methods： Healthy male Wistar rats were randomly divided into Control group,Model group,Adriamycin group,Ganshu-point（BL18） group,Zusanli-point（ST36） group.Liver cancer models were prepared by injection of diethylnitrosamine.The expression of Wnt3a and Wnt10b was detected by RT-PCR and the structure and morphological changes of hepatocytes were observed.Results：Compared with the control group,the expression of Wnt10b and Wnt3a in Model group,Zusanli-point（ST36） group,Ganshu-point（BL18） group and Adriamycin group increased significantly; Compared with the Model group,Adriamycin group,Ganshu-point（BL18） group and Zusanli-point（ST36） group expression reduced; Compared with Adriamycin group,the expression of Zusanli-point（ST36） group and Ganshu-point（BL18） group decreased obviously; Compared with the Zusanli-point（ST36） group,Ganshu-point（BL18） group expression was significantly increased.Visible were observed：Control group of normal liver tissue; The cells in the model group had false lobular formation; Adriamycin group was significantly hyperplastic lesions; In the Ganshu-point（BL18） group,the nucleus became more pleomorphic and the nuclear plasma fraction disappeared; Fibrous changes in the Zusanli-point（ST36） group.Conclusions： Scarring moxibustion Ganshu-point（BL18）,Zusanli-point（ST36） for hepatic carcinomain rats have therapeutic effects.The mechanism may be by regulating the Wnt/β-catenin signaling pathway Wnt3a,Wnt10b expression to achieve.
