新型动物精子上游分离器的研究

摘要 本研究旨在为动物体外受精提供具有活力的精子,运用新型精子上游分离器筛选出高活性的精子。本实验应用3、4.5、6 mm 3种不同上浮高度的精子分离器对猪精子进行筛选,并检测3种不同高度精子分离器对分离猪精子质量的影响,筛选出最理想的筛选高度的精子分离器。结果表明:上浮高度为4.5 mm精子分离器的精子活率(85.11%)显著优于筛选高度为3 mm的分离器(74.60%)和普通上游法(62.83%)(P<0.05),但与筛选高度为6 mm的分离器差异不显著(P>0.05);4.5 mm精子分离器的畸形率(7.49%)显著低于对照组和3 mm的分离器(P<0.05);4.5 mm精子分离器的精子直线速度和路径速度最高(P<0.05),但分离精子的密度较普通上游法要低;各组之间猪精子其他质量参数差异不显著(P>0.05)。综合可知,上游高度为4.5 mm的精子分离器优于其他高度的分离器及普通上游法,应用上游高度为4.5 mm的精子分离器能够分离出具有活力的猪精子。 In order to isolate high motility sperm for in vitro fertilization, to screen out the highly effective selection method of viable sperm. In this study, we used three different sperm separators (3, 4.5, 6 mm) to isolate high motility sperm, and test that the effect of three different sperm separators on quality of separated boar sperm. The results showed that 4.5 mm sperm separator in sperm motility (85.11%) was significantly better than the 3 mm sperm separator (74.60%) and (62.83%) swim- up method (P〈0.05), but no difference between 6 mm and 4.5 mm sperm separator (P〉0.05); However, malformation rate of 4.5 mm sperm separator (7.49%) was significantly lower than that of the other groups (P〈0.05). In addition, 4.5 mm sperm separator in sperm VSL and VAP in all groups was the highest (P〈0.05), but the separation of sperm density is lower than swim-up group; In the other quality parameters, there were no significant difference among the groups (P〉0.05). Above the results showed that the sperm separator with a height of 4.5 swim-up method, and it could separate viable sperm for IVF.
