高通量测序分析酵母培养物对肉仔鸡盲肠菌群的影响 被引量:5

Effect of Different Fermentation Duration of Yeast(Saccharomyces cerevisiae) Culture on the Intestinal Microbiota by Miseq High-throughput Sequencing

摘要 本实验旨在探讨酵母培养物对肉仔鸡盲肠菌群的影响。选取336羽健康且体重相近的1日龄AA肉仔鸡,随机分7个处理组,每组6个重复,每个重复8只。A组(对照组)饲喂基础日粮,B、C、D、E、F组分别在基础日粮中添加发酵时间为12、24、36、48、60 h的酵母培养物,G组在基础日粮中添加"达农威益康V XP"酵母培养物产品。预试期7 d,正试期42 d。采用Illumina Miseq高通量测序技术对肉仔鸡盲肠内容物菌群分析。结果表明:与A组(对照组)相比,C、D组肉仔鸡的平均日增重显著提高(P<0.05),耗料增重比显著降低(P<0.05);而D组肉仔鸡盲肠菌群物种多样性最高,C组最低;在Phylum(门)的水平上,各组中的Firmicutes(厚壁菌门)比例最高,而C组样品中Verrucomicrobia(疣微菌门)含量较其他组显著增加;在Genus(属)的水平上,7组样品的盲肠内容物中以Bacteroides、Faecalibacterium、Lactobacillus、Blautia等菌属为主。综上,不同发酵时间的酵母培养物对肠道菌群有显著影响,可通过调整发酵时间来控制其代谢产物,进而影响动物的生产性能。 This study was conducted to investigate the effect of different fermentation time of Yeast (Saccharomyces cerevisiae) Culture on the intestinal microbiota. One-day-old AA 336 broilers with the comparable relative quality were randomly assigned to 7 groups with 6 replicates of 8 broilers each. The broilers in the control treatment A were only fed a basal diet ,and treatment B, C, D, E, F were yeast culture groups, supplemented with fermentation time at 12 h, 24 h, 36 h, 48 h and 60 h of yeast culture,and treatment G was Diamond V XP, respectively. The experiment was based on 42 d to 7 d preliminary trial period. After 42 d. Cecum contents were collected and the intestinal microbiota was detected by Miseq high-throughput sequencing technology. The results showed as follow: Compared with the control group, at 7-42 d, averaged daily gain (ADG) in C and D group were significantly higher (P〈0.05), and that two groups of feed to gain ratio(F/G)were significantly lower. D group had the highest intestinal microbiota diversity while C group, exhibited the lowest species diversity. In the phylum level, Firmicutes was the most abundant bacterial in 7 groups, while the proportion of Verrucomicrobia was higher in C group was against the other group. In the genus level, Bacteroides, Faecalibacterium, lactobacillus, Blautia were the more abundant bacterial in 7 groups. In conclusion, different time of fermentation of yeast culture has a significant effect on intestinal flora, and thus we can control composition metabolites of yeast culture by adjusting the fermentation time, in turn, adjusting the structure of intestinal flora, affect the production performance of animal.
