三七皂苷对冈田酸拟AD细胞模型的作用 被引量:1

Effects of Panax notoginseng saponins on cellular model of alzhemier disease induced by okadaic acid

摘要 目的利用蛋白磷酸酶抑制剂冈田酸模拟AD病细胞损伤模型,探讨三七总皂苷的抗AD作用及其可能的作用机制。方法三七总皂苷提取物与PC12细胞预孵育24 h,利用终浓度为25 nmol/L OA处理PC12细胞,采用JC-1荧光检测法测定线粒体膜电位、Eura-2/AM荧光标记法测定胞内Ca^(2+)浓度、Caspase-3酶活检测试剂盒检测,Caspase-3的活性,同时用Western blot检测Bcl-2、Bax、Caspase-3的表达情况。结果三七总苷能提高AD细胞模型的线粒体膜电位,降低胞内Ca^(2+)浓度;减少Bax和Caspase-3的表达,提高Bcl-2的表达。结论三七总皂苷对OA引起的神经细胞凋亡具有一定的保护作用。 Objective The anti - AD effect of Panax notoginseng saponins and its possible mechanism were investigated by using the protein phosphatase inhibitor okadaic acid to induce the AD cell injury model. Methods Cultured PC12 cells were treated with 25 nmol/L OA for 24 h at the presence of Panax notoginseng saponins, JC - 1 fluorescence, intracellular free Ca2+ concentration ( [ Ca2+ ] i) and the activity of Caspase 3were determined by assay kit respectively. The expression of Bcl -2, Bax and Caspase 3 was detected by Western blot. Results The results showed that Panax notoginseng can increase the mitochondrial membrane po- tential of AD cell model, decrease the intracellular Ca2+ concentration. And also can reduce the expression of Bax and Caspase - 3 and increase the expression of Bcl -2. Conclusion The results showed that Panax notoginseng saponins had a protective effect on PC12 cell injury induced by OA.
