摘要
目的研究鹿筋胶原蛋白中氨基酸组成及其对骨细胞的作用。方法以双酶法制备的鹿筋胶原蛋白多肽为研究对象,利用柱前衍生化的方法测定其氨基酸的相对含量;采用MTT法检测其对C518软骨细胞和MC3T3-E1成骨细胞的增殖作用。结果在鹿筋胶原蛋白中,甘氨酸相对含量最高,为31.81%,羟脯氨酸的相对含量为6.99%,符合胶原蛋白组成的特点。与对照组比较,鹿筋胶原蛋白多肽对C518细胞、MC3T3-E1细胞均具有显著的增殖作用,其增殖作用呈量效关系,当浓度为200μg/m L时,增殖作用最为显著。结论鹿筋胶原蛋白多肽对骨细胞具有明显的增殖作用,为其产品的开发提供理论依据。
Objective To research the amino acid composition of deer sinew collagen peptide and its effect on osteocytes. Meth- ods The collagen peptide prepared by double enzyme method was used as the research object. The relative content of amino acids was determined by precolumn derivatization. The proliferation of C518 chondrocytes and MC3T3 -E1 osteoblasts was detected by MTT assay. Results In the collagen peptide, the relative content of glycine was the highest, accounted for 31.81% , and the rel- ative content of hydroxyproline was 6.99% , which were consistent with the composition of collagen. Compared with the control group, collagen peptide had significant proliferation effect on C518 cells and MC3T3 -E1 cells, and its proliferation was dose- dependent. When the concentration was 200 μg/mL, the proliferation was the most significant. Conclusion The collagen peptide has obvious proliferative effect on osteocytes, and provides theoretical basis for the development of its products.
