摘要
目的观察雌激素(E_2)缺乏状态下年轻与老龄化C57BL/6N小鼠脾脏IL-2、6、10和TNF-α水平的变化。方法清洁级健康雌性C57BL/6小鼠80只,分为手术去卵巢组、药物去卵巢组、自然绝经组及发情间期组(对照组),经Con-A刺激后RT-PCR技术检测4组小鼠脾脏IL-2、6、10和TNF-α水平,ELISA检测血清E_2水平证实分组可靠性。结果 3组实验组血清E_2水平均明显低于对照组(P<0.01),而E_2水平在3组实验组内比较差异无统计学意义。小鼠脾脏表达IL-2、IL-6和TNF-α水平在自然绝经组升高,与其他3组比较差异均有统计学意义(P<0.05);而3组实验组IL-10水平略低于对照组,差异无统计学意义(P>0.05);然而,年轻组别中(发情间期组、手术去卵巢组、药物去卵巢组)未发现有IL-2、IL-6、IL-10和TNF-αmRNA表达的差异。结论 Con-A刺激后E_2缺乏状态下年轻与老龄化C57BL/6N小鼠之间细胞因子模式是不同的,E_2缺乏对IL-6、IL-2和TNF-α水平影响作用有限。
Objective To compare the levels of IL-2, IL-6,IL-10 and TNF-α between young and aged C57BL/6N mice with the common feature of low E2 status.Methods Twenty natural aged, 20 surgical variectomy and 20 medical variectomy of C57BL/6N mice were chosen in our studies. 20 stage of diestrus served as controls. Blood and spleens were collected 4 hours after injection of con-A. Reverse transcription-pelymerase chain reaction (RT-PCR) was used to determine the mRNA levels of IL-2, IL-6, IL-10 and TNF-α in spleen cells. Serum E2 levels were assessed using enzyme linked immunosorbent assay (ELISA).Results The serum E2 level of three experiment groups was significantly lower than that of control group (P〈0.01), while the difference was not significant when compared it within experiment groups. The levels of IL-2, IL-6 and TNF-α were increased in aged group, and the difference reached statistically significant when compared them with any other groups(P〈0.05). While the IL-10 level was slight decreased in three experiment groups compared with control group (P〉0.05).Conclusion The cytokines response to Con-A stimulus between young and aged C57BL/6N mice with estradiol deficiency was different. The effect of E2 to these cytokines may be marginal.
