摘要
目的:观察黄芪解毒汤对人乳腺癌细胞MCF-7凋亡及Notch1基因表达的影响,探讨该方抑制乳腺癌复发转移的可能机制。方法:制备含药血清,分为黄芪解毒汤血清组、阿霉素血清组、参一胶囊血清组,另设生理盐水血清组。常规培养细胞,MTT法检测不同浓度含药血清对MCF-7细胞增殖的影响;选择最佳抑制浓度的含药血清分别干预乳腺癌细胞,干预3 d后流式细胞仪检测各组MCF-7细胞凋亡率,实时荧光定量PCR检测各组MCF-7细胞Notch1基因的表达。结果:MTT法检测结果显示,3种含药血清均可抑制MCF-7细胞增殖(P<0.05)且对细胞抑制最佳浓度为20%含药血清。流式细胞检测结果显示,与生理盐水血清组比较,黄芪解毒汤血清组、参一胶囊血清组、阿霉素血清组均可明显诱导MCF-7细胞凋亡(P<0.05);实时荧光定量PCR结果显示,与生理盐水血清组比较,黄芪解毒汤血清组、参一胶囊血清组、阿霉素血清组Notch1基因表达均明显降低(P<0.05)。结论:黄芪解毒汤可能通过诱导MCF-7凋亡、降低Notch1基因表达起到抑制乳腺癌复发转移的作用。
Objective: To observe the effects of Huangqi Jiedu Decoction on the apoptosis of human breast cancer cells line MCF-7 and the mRNA expression of Notchl ,and discuss its possible mechanism of inhibiting recurrence and metastasis of breast cancer. Methods: The drug medicated serum was prepared. The cells were divided into the Huangqi Jiedu Decoction group, adriamycin group, Shenyi Capsule group. In addition, the ceils intervened by serum from rats treated with normal saline were taken as the normal saline group. The cells were routinely cultured. The effects of drug medicated serum with different concentrations on the cell proliferation of MCF-7 were detected by MTI" assay. The breast cancer cells were intervened by different drug medicated serum at the optimum inhibitory concentration. Three days later,the apoptosis rate of MCF-7 cells in each group was detected by flow cytometry,and the Notch1 mRNA expression of MCF-7 ceils in each group was detected by real time fluorescent quantitative PCR. Results: MTT assay showed that the three kinds of drug medicated serum could all inhibit the cell proliferation of MCF-7 ( P 〈0. 05 ), and the drug medicated serum at 20% was the optimum concentration. The results of flow cytometry showed that compared with the normal saline group, the drug medicated serum of Huangqi Jiedu Decoction, adriamycin and Shenyi Capsule could obviously induce the apoptosis of MCF- 7 ceils ( P 〈 0.05 ). The results of real time fluorescent quantitative PCR showed that compared with the normal saline group, the Notch1 mRNA expression was obviously decreased in the Huangqi Jiedu Decoction group, adriamycin group and Shenyi Capsule group ( P 〈 0.05 ). Conclusion : Huangqi Jiedu Decoction may inhibit the recurrence and metastasis of breast cancer by inducing the apoptosis of MCF-7 cells and decreasing the Notch1 mRNA expression.
