摘要
本试验通过在含有0.05%琼脂粉的沙氏葡萄糖肉汤培养基中培养Duddingtonia flagrans(D.flagrans)再转接到玉米粒培养基的方法,对D.flagrans厚壁孢子进行培养,之后将其洗脱并制备成冻干制剂,然后对冻干制剂的萌发率和杀虫率进行观察。结果显示,D.flagrans冻干制剂厚壁孢子在接种玉米粉琼脂培养基(CMA)培养后5d萌发率达到峰值78.3%;经口饲喂绵羊D.flagrans冻干厚壁孢子剂量为1×106个/kg时,幼虫平均减少率为81.4%。结果表明,D.flagrans冻干制剂使用方便,复苏和杀虫效果较好,其在生物防制家畜胃肠道线虫病方面有很大的应用前景。
In order to obtain the germination rate and trapping fungus-Duddingtonia flagrans ( D.flagrans dextrose broth medium with 0. 05% agar, and then insecticidal effect ),the fungus first transferred to the of freeze-dried incubated in corn medium. sabouraud After cultured,the fungi were chlamydospores from media eluted and lyophilized. The results showed that the maximum germination rate of lyophilized D.flagrans chlamydospores was 78.3% on the 5th day after inoculation; the average percent reduction of nematodes larves was 81.4% when the sheep were feed by freeze-dry preparation at the dose of 10%; fungal spores/kg body weight. The results indicated that the freeze-dried D. flagrans production is not only convenient to use but also has good effect on recovering and killing nematode. Proved by the trials, this lyophilized prepara tion of D. jlagrans has great application prospects in biological control of livestock gastrointestinal nematodes.
