摘要
为了研究维药阿里红各活性成分抗鸡新城疫病毒(NDV)的活性,本试验分别提取了阿里红多糖、多酚、黄酮、生物碱、挥发油并测定其含量。通过MTT法测定阿里红各活性成分对鸡胚成纤维细胞的最大安全浓度。以3种加药方式将药物与NDV加入已制备好的鸡胚成纤维细胞上,即先加药后接种病毒,先接种病毒后加药,病毒和药物混合作用后加入,以检测各组分对NDV的阻断作用、抑制作用以及直接杀灭作用。结果表明,多糖、多酚、黄酮、生物碱、挥发油均有抗病毒活性,其中多糖对NDV的直接杀灭作用及抑制作用强于对NDV的阻断作用。多糖、多酚、黄酮、生物碱、挥发油在抑制作用下的病毒抑制率分别为49.12%,39.64%,42.45%,40.00%,18.59%;杀灭作用下的病毒抑制率分别为45.35%,35.71%,38.57%,35.71%,41.07%;阻断作用下的病毒抑制率分别为15.58%,8.57%,32.98%,43.11%,31.42%。综上可知,维药阿里红的活性成分多糖和生物碱的抗NDV活性较好,可以作为进一步研究的材料。
To research the antiviral effects of five activity ingredients from Fomes officinalis on Newcastle disease virus (NDV). This experiment extracted the Fom es officinalis polysaccharides, polyphenols, flavonoids, alkaloids, volatile oil, respectively and determinated their content. The safe concentration and growth of chick embryo fibroblast (CEF) were assayed by MTT method. Under the safety range of concentration, the block virus activity, anti virus activity and virus-killing activ ity of polysaccharide,polyphenol, flavone,aikaloid, essential oil were detected through the way of pre-adding drug, post-adding drug, adding drug with NDV. After culture 72 hours, observed the CEF lesion severity,and measuring the D490 value of cell by MTT method,evaluated the influence of the drug on NDV infection ability to the cells. The results showed thai all ingredients had antiviral activity. The direct killing and inhibition effect of polysaccharide on NDV was better than blocking effect. Under the inhibition pattern, virus inhibition rate of polysaccharide, polyphenol, flavone, alkaloid, essential oil were 49.12%, 39.64%, 42.45%, 40.00%, 18.59%, respectively ; under the action pattern, killing virus inhibition rate were 45. 35%,35. 71%,38. 57%,35. 71%,43.11%, 31.42%, respectively. In summary, the antiviral activity of potysaccharide and alkaloid in this five ingredients were better than the others and would be as the candidate materials for further research.
