摘要
旨在研究中国荷斯坦牛SLC35A3-66基因SNP突变与疾病及泌乳性能的相关性,为加快奶牛的育种进程提供理论基础。通过对866头中国荷斯坦牛SLC35A3基因编码区进行直接测序和飞行质谱分析,检测SLC35A3基因DNA序列第559位上核苷酸突变G>T,同时收集采样牛只疾病与泌乳性能等信息,用Logistic回归分析其与流产的相关关系,用方差分析法分析SLC35A3-66G>T突变与各胎次临床乳房炎病例数和蹄病发生次数、生产寿命及泌乳性能的相关关系,用Cox回归分析不同基因型奶牛的生存曲线。结果表明:中国荷斯坦牛SLC35A3基因编码区SNP存在1个SNP突变位点c.66G>T;SLC35A3-66G>T与乳脂率、蛋白率、脂蛋白比和总固体极显著性相关(P<0.01),与体细胞评分(SCS)和乳糖含量显著性相关(P<0.05),GT型个体各项指标均显著高于GG型个体;SLC35A3-66G>T与各胎次流产、临床乳房炎发生次数和蹄病发生次数之间无显著性相关(P>0.05);SLC35A3-66G>T与离群月龄显著性相关(P<0.05),GT型个体的离群月龄显著高于GG型个体,与生产月龄和离群胎次无显著性相关(P>0.05)。Cox回归分析表明,SLC35A3-66G>T与荷斯坦牛生产寿命显著性相关(P<0.05),GT型个体的离群月龄显著高于GG型纯合子个体;SLC35A3-66G>T与乳脂率、蛋白率、脂蛋白比和总固体极显著性相关(P<0.01),与乳糖、SCS和奶牛生产寿命显著性相关(P<0.05)。该结果为中国荷斯坦牛泌乳性能和乳房炎抗性的分子标记辅助选择提供参考。
To explore the correlation between SL(735A3-66 G〉T and the disease and milking traits of Chinese Holstein cows,and to provide the basis for the improvement of breeding. The mutation G〉T at 559th in coding region of SLC35A3 gene was detected by directly sequencing and flight mass spectrometry in 866 Chinese Holstein cows from a large dairy farm. The milking traits,clinical mastitis and lameness of tested cows were collected from the management system of dairy farm. The correlations between SLC35A3 66 G〉T and the abortion,disease and milking traits of Chinese Holstein cows were analyzed using Logistic regression, multivariate analysis of variance and Cox regression analysis. The results showed that:only SLC35A3-66 G2〉T mutation was found in the 559th position of SLC35A3 gene nucleotide sequence;the SLC35A3-66 G〉T was extremely significantly related to the FC,PC,FPR and TS(P〈0.01) and significantly related to SCS and LC (P〈0. 05). The FC, PC, FPR, TS and LC for the individuals with GT genotypes were higher than GG genotypes significantly, and the SCS for the individuals with GT genotypes was lower than GG genotypes. SLC35A3-66 G〉T was not related to abortion,the number of dairy cows suffered from clinical mastitis and lameness(P〉0.05). The SLC35A3-66 G〉T was significantly related to culling month of cows(P〈0.05),and the culling month for individual with GT genotype was higher than that of GG genotype(P〈0.05). SLC35A3-66 G〉T was not related to productive months and culling parity of cows. Cox regression analysis showed that SLC35A3-66 G〉T was significantly related to productive life(P〈0. 05) ,the culling months for individuals with GT genotype were significantly higher than that with GG genotype. Conclusion:SLC35A3-66 G〉 T had extremely significant correlation with FC,PC,FPR and TS(P〈0.01) and significant correlation with LC,SCS and productive lifetime(P〈0.05). These results provided basis for the molecular marker assisted selection of milking traits and mastitis resistance in Chinese Holstein cows.
