摘要
目的探索长链非编码RNA-H19对急性髓系白血病细胞增殖和细胞凋亡的调控作用。方法采用RAN干扰技术对急性髓系白血病细胞系HL-60、THP-1和U937的LncRNA-H19进行表达沉默,用实时荧光定量PCR(qPCR)检测LncRNA-H19在细胞中的表达情况。用MTT检测细胞增殖情况,用流式细胞术检测细胞凋亡情况。结果三种白血病细胞系分别被3条si-RNA干扰后,LncRNA-H19表达均明显下调。MTT结果显示,RNA干扰后,LncRNA-H19干扰细胞组的细胞增殖明显低于阴性对照组,而流式细胞术显示,siH19干扰细胞组的细胞凋亡明显高于阴性对照组,差异有统计学意义(P<0.05)。结论急性髓系白血病细胞中,LncRNA-H19具有调控急性髓系白血病细胞增殖和细胞凋亡的功能。
Objective To explore the regulating effect of long non-coding RNA-H19" LncRNA- H19) on cell proliferation and apoptosis in different acute myeloid leukemia cell lines. Methods HL-60, THP-1 and U937 cellswere transfected with si-H19 and si- NC, respectively. MTT assay was performed to determine the cell apoptosis was assayed by flow cytometry using Annexin V- FITC/PI. Results The expression AML cells was significantly down-regulated after the treatment of si-H19 for 72 hours. Down-regulation of LncRNA-H19 in AML cell lines HL-0, THP-1 and U937 inhibited cell proliferation,and induced cell apoptosisConclusion LncRNA- H19 can regulate cell proliferation and apoptosis in acute myeloid leukemia cell lines.
