针刺“神门”穴对睡眠剥夺大鼠脑电波及其认知能力的影响 被引量:23

Effects of Acupuncture at“Shenmen”(HT 7) on Brainwaves and Cognitive Ability in Rats with Sleep Deprivation

摘要 目的:探讨"神门"穴对睡眠剥夺大鼠脑电波及其认知能力的影响。方法:SD大鼠随机分为空白组、模型组、针刺组和假针刺组,每组15只。采用睡眠剥夺法制备大鼠失眠模型。针刺组取"神门"穴针刺,1次/d,20min/次,共7d;假针刺组仅刺入"神门"穴皮肤1mm,不留针,1次/d,共7d。采用六角迷宫箱评价大鼠学习记忆能力,BL-420F生理记录仪于实验第1、4、7和10天对所有大鼠分别进行30min脑电图记录,分析并记录各波频次。结果:睡眠剥夺后的模型组大鼠寻找出口频率明显增加,寻找时间缩短,寻找错误次数明显增加,认知率降低(P<0.01);与模型组和假针刺组比较,针刺组大鼠寻找频率明显降低,寻找时间较长,寻找错误次数减少,认知率升高(P<0.05)。与空白组比较,模型组脑电图δ波显著减少,β波显著增多(P<0.01);与模型组和假针刺组相比,针刺组δ波显著增多,β波显著减少(P<0.05)。假针刺组和模型组各指标比较差异均无统计学意义(P>0.05)。结论:针刺"神门"穴能有效改善睡眠剥夺后大鼠行为认知能力和脑电波。 Objective To observe the effects of acupuncture at "Shenmen" (HT 7) on brainwaves and cognitive ability in rats with sleep deprivation. Methods SD rats were randomly divided into control group, model group, acupuncture group and sham acupuncture group, 15 rats in each group. Insomnia model rats were established by sleep deprivation for 72 hours. Acupuncture was applied to acupuncture group at bilateral "Shenmen" (HT 7) for 20 min, once daily for 7 days. Rats in the sham acupuncture group received superficial insertion of 1 mm and without retaining needle. The learning and memory abilities of rats were evaluated by Hexagonal maze. The BL-420 F physiological recorder was used to record EEG for 30 rain on the 1st , 4th , 7t" and 10th day respectively, and the frequency of each wave was analyzed. Resu|ts After sleep deprivation, the frequency of searching the exits of Hexagonal maze in the model group was obviously increased, the searching time was shortened, the number of searching errors was increased obviously, and the cognitive rate was decreased (P〈0.01). Compared with the model group and the sham acupuncture group, the frequency of searching the exits in the acupuncture group was obviously decreased, the searching time was longer, the number of seeking errors was decreased, and the cognitive rate was increased (P〈0.05). Compared with the control group, the EEG of 6 wave in the model group was decreased while ~ wave increased with statistical significance (P〈 0.01). Compared with the model group, the 6 wave of rats in the acupuncture group was significantly increased and the β wave was significantly decreased (P〈0.05). There were no statistically significant differences between the sham acupuncture group and the model group ( P〈0.05), Conclusion Acupuncture at HT 7 can effectively improve the cognitive abilities and brainwaves in sleep deprived rats.
