MAP kinase级联信号在植物生长发育、抗病和抗逆等进程中发挥重要作用。水稻OsMKK10-2基因受脱落酸、干旱、盐和冷胁迫诱导表达。酵母双杂和pull down实验表明OsMKK10-2能与下游激酶OsMPK1、OsMPK5和OsMPK6相互作用,且OsMKK10-2N端MPK停泊位点决定了其与下游MPKs间的互作。OsMKK10-2:GFP融合蛋白定位在烟草细胞质和细胞核。不过,较长时间表达OsMKK10-2可以导致烟草细胞死亡和ROS产生,且依赖于其N端的MPK停泊位点。研究结果为进一步揭示OsMKK10-2的生物学功能奠定了基础。
MAP kinase cascades play vital roles in plant growth and development, disease resistance and stress tolerance. The expression of OsMKKIO-2 gene in rice seedlings was induced by ABA, drought, salt, and cold treatments. Yeast two hybrid and pull-down assays revealed that OsMKK10-2 interacted with OsMPK1, OsMPK5 and OsMPK6, and that its N-terminal MPK-docking site was essential for the interaction with these MPKs. The OsMKK10-2:GFP fusion protein was shown to target the nucleus and cytoplasm of tobacco plant cells. Moreover, transient overexpression of OsMKK10-2 in tobacco induced cell death and H202 production, which required its MPK docking site. Our results provide a foundation for further understanding the biological function of OsMKK10-2.
