摘要 探讨纺织品中快速检测沙门氏菌的方法。应用了环介导等温扩增技术结合基质辅助激光解吸电离飞行时间质谱技术建立纺织品中沙门氏菌的快速检测方法。沙门氏菌invA基因序列具有致病性菌株属特异性,根据其特点设计出一套沙门氏菌环介导等温引物。环介导等温扩增结果为可疑或阳性时,分离菌株,通过基质辅助技术快速鉴定结果。选择典型参考菌株做特异性试验;沙门氏菌菌液稀释成不同梯度做灵敏度试验。试验结果表明:该方法特异性好,灵敏度较高,检测限可达4.6CFU/mL。认为:该方法可用于快速检测纺织品中沙门氏菌。 The methods for the rapid detection of salmonella in textiles were discussed. The rapid detection method for salmonella in textiles was established by applying loop-mediated isothermal amplification technology and combining with matrix-assisted laser desorption Ionization time-of-flight mass spectrometry technology. The invA gene order for salmonella had the specificity of pathogenic bacterial strain category. A set of salmonella LAMP primers were designed according to its properties. When the amplification effect of LAMP was suspicious or positive,the strain was separated. Through MALDI TOF MS technology, the results were rapidly identified. The classic referred strain was chosen for specificity test. The salmonella bacteria solutions were diluted into different gradients for sensitivity test. The test results show that the method has better specificity and sensitivity. The test limitation can reach 4.5 CFU/mL. It is considered that the method can be used for rapid detection of sal- monella in textiles.
