公鸡鸡冠发育相关候选基因的表达分析 被引量:7

摘要 为了探索鸡冠发育的分子机制,利用实时荧光定量PCR技术(qRT-PCR)对前期采用转录组测序技术筛选出的7个与鸡冠发育相关的候选基因(BMP2、TPPP3、HSD17B2、CYP2W1、CYP1A4、EDA2R、HAL)进行表达分析。选取200只5周龄青脚麻鸡A系公鸡,从中筛选出15只高鸡冠和15只矮鸡冠公鸡,采用qRT-PCR定量分析7个候选基因在两组个体鸡冠组织的相对表达量,同时检测了7个候选基因在心、肝等11个组织的表达谱。结果显示:CYP2W1和TPPP3在高冠组表达量极显著高于矮冠组(P<0.01);CYP4A1和EDA2R在高冠组表达量高于矮冠组(P<0.05);HAL在高冠组中表达量极显著低于矮冠组(P<0.01);HSD17B2和BMP2在高冠组中表达量显著低于矮冠组(P<0.05)。表明BMP2、TPPP3、CYP2W1、EDA2R四个基因在鸡冠中的表达属于较高水平表达,其余基因在鸡冠组织中属于中等水平表达。提示这7个候选基因可能在调控鸡冠发育中发挥重要作用。 Abstract: To explore the molecular mechanism of cockscomb development, 7 candidate genes (BMP2, TPPP3, HSD17B2, CYP2W1, CYPIA4, EDA2R, HAL) selected from the previous transeriptome sequencing were analyzed by qRT-PCR. 15 high-cockscomb rooters and 15 low-cockscomb rooters were selected from 200 partridge-shank roosters (line A)aged 5 weeks. The relative expression of seven candidate genes in comb of those roosters was quantitatively analyzed by qRT-PCR, meanwhile, the tissue expression profile of seven candidate genes was quantitatively analyzed in 11 tissues which were heart, liver and so on. The results showed that 7 candidate genes were all expressed in cockscomb, CYP2W1 and TPPP3 were extremely significantly expressed in high-cockscomb group compared with low-cockscomb group(P〈0.01), meanwhile, CYP4A1 and EDA2R were significantly expressed in high-cockscomb group (P〈0.05). However, HAL was extremely significantly expressed in low-cockscomb group compared with high-cockscomb group (P〈0.01),meanwhile, HSD17B2 and BMP significantly expressed in low-cockscomb group (P〈0.05). The results of tissue expression profile showed that the expression of BMP2, TPPP3, CYP2W1 and EDA2R genes in the cockscomb was high, and the other genes were expressed at the middle level in the cockscomb. The results suggested that seven candidate genes may play an important role in regulating the development of cockscomb.
