冬凌草甲素对人脑胶质瘤细胞株U251杀伤、增殖及NF-κB蛋白表达的影响 被引量:2

摘要 目的观察冬凌草甲素对人脑胶质瘤细胞株U251杀伤、增殖及NF-κB蛋白表达的影响。方法将浓度为10、20、40、80μmol/L冬凌草甲素与U251细胞(实验组)分别孵育,同时设不加冬凌草甲素培养的U251细胞作对照组,分别于不同时间点计数U251、计算冬凌草甲素对U251细胞的IC50、测算U251细胞增殖抑制率、检测U251细胞中的NF-κB蛋白。结果与对照组相比,实验组加入40、80μmol/L的冬凌草甲素后U251细胞数明显减少(P均<0.05)。冬凌草甲素作用24、48、72 h时对U251细胞的IC50分别为82.2、65.3、44.1μmol/L。与20μmol/L冬凌草甲素相比,40、80μmol/L冬凌草甲素作用48、72 h时的细胞增殖抑制率明显降低(P均<0.05)。与对照组相比,40、80μmol/L的冬凌草甲素作用48、72 h时,U251细胞中NF-κB蛋白的相对表达量明显降低(P均<0.05)。结论冬凌草甲素在体外可杀伤人脑胶质瘤细胞株U251,抑制其增殖,下调其NF-κB蛋白的表达;冬凌草甲素对U251细胞的杀伤、增殖抑制作用可与其对NF-κB信号通路发挥调节作用有关。 Objective To observe effects of oridonin on killing,proliferation,and expression of NF-κB protein of human glioma U251 cell line. Methods U251 cells were incubated with oridonin at the concentrations of 10,20,40,and80 μmol/L respectively. At the same time,U251 cells incubated without oridonin were taken as the control group. U251 counting,IC50,U251 cell proliferation inhibition rate and expression of NF-κB were detected at different time points. Results Compared with the control group,the number of U251 cells in the 40 and 80 μmol/L oridonin groups decreased significantly( P 〈0. 05). After incubation for 24,48 and 72 h,the IC50 of U251 cells was 82. 2,65. 3 and 44. 1 μmol/L,respectively. Compared with 20 μmol/L oridonin group,the cell proliferation inhibition rates of the 40 and 80 μmol/L oridonin groups were significantly reduced at 48 and 72 h( P〈 0. 05). Compared with the control group,the expression of NF-κB in U251 cells decreased significantly( P 〈 0. 05) in the 40 and 80 μmol/L oridonin group at 48 and 72 h. Conclusions Oridonin can kill U251 cells,inhibit its proliferation and down-regulate NF-κB protein in vitro. The killing effect and inhibition can be related to the regulation of NF-κB signaling pathway.
