交泰丸等4首不同方剂对2型糖尿病大鼠治疗作用的比较研究 被引量:2

摘要 目的:比较白虎加人参汤(BHJRST)、黄连解毒汤(HLJDT)、六味地黄丸(LWDHW)和交泰丸(JTW)对高糖高脂饲料喂养联合小剂量链脲佐菌素(STZ)致2型糖尿病大鼠模型的药效作用。方法:4周龄SD大鼠高糖高脂饲料喂养5周后腹腔注射小剂量STZ(40 mg/kg)制备2型糖尿病模型,分为BHJRST、HLJDT组、LWDHW组和JTW组、模型组,并设正常组作对照,连续灌胃21 d。记录各组大鼠饮食量、体质量、空腹血糖(FBG)的动态变化,实验末检测血清游离脂肪酸(free fatty acid,FFA)、三酰甘油(triglyceride,TG)、总胆固醇(total cholesterol,TC)、胰岛素(insulin,INS)的变化,计算胰岛素敏感指数(insulin sensitivity index,ISI),并观察胰岛组织病理与超微结构变化。结果:与正常组比较,模型组体质量明显下降,饮水量、摄食量增加(P<0.01),FBG和FFA、TG升高(P<0.05),INS与ISI降低(P<0.05),形态学显示胰岛及β细胞结构严重破坏。与模型组比较,JTW组FBG、FFA、TG水平明显降低(P<0.05),ISI明显升高(P<0.05),胰岛结构及β细胞形态改善;而LWDHW组饮水、进食量减少(P<0.05),血清FFA明显降低(P<0.05),ISI明显升高(P<0.01),胰岛β细胞形态亦有所改善。结论:JTW、LWDHW对2型糖尿病大鼠模型具有明显保护作用,不同方剂对同一疾病可能存在不同的作用特点和机制。 Objective:To compare the therapeutic effects of four traditional Chinese medicine prescriptions Baihu Jiarenshen Tang,Huanglian Jiedu Tang,Liuwei Dihuang Wan and Jiaotai Wan on type 2 diabetes rat model induced by a high-fat,high-sucrose diet and low dose of streptozotoxin(STZ).Methods:Male Sprague-Dawley rats were fed with regular chow or high-fat,high-sucrose diet for 5 weeks and then injected with vehicle or streptozotocin(STZ) 40 mg/kg respectively.After fasting blood glucose(FBG) was measured to ensure type 2 diabetes happened,the model rats were randomly divided to treat with Baihu Jiarenshen Tang,Huanglian Jiedu Tang,Liuwei Dihuang Wan and Jiaotai Wan or vehicle orally for 3 weeks.Body weight,food and water intake,and FBG were monitored weekly throughout the study,serum free fatty acids(free fatty acid,FFA),triglyceride(triglyceride,TG),total cholesterol(total cholesterol,TC) and insulin(insulin,INS) were measured and insulin sensitivity index(insulin sensitivity index,ISI) were calculated when rats were sacrificed.Pancreas of each animal were resected and partially sectioned for morphological study.Results:Compared with the normal group,the type 2 diabetes rats had the symptoms of polydipsia,polyphagia and weight loss(P〈0.0);the FBG,TG,FFA levels were significantly higher while the INS and ISI were reduced in model rats(P〈0.05);the changes in islet morphology of model rats included loss of definition of the islet boundary,reduction ofβ-cell mass,depletion of insulin-containing secretory granules in β-cells.Compared with model rats,the FBG,TG and FFA level were decreased significantly and ISI was increased in Jiaotai Wan group after 3 weeks of administration(P〈0.05),the islet morphology of Jiaotai Wan group exhibited better islet integrity,increased proportion ofβ-cells and greaterβ-cell granulations.The group treated with Liuwei Dihuang Wan had fewer water intake and food cosumption at the end of 3 weeks of administration(P〈0.05),serum FFA level was decreased and ISI was increased significantly(P〈0.05,P〈0.01),and the islet morphology of Liuwei Dihuang Wan group was similar to Jiaotai Wan group.Conclusion:Jiaotai Wan and Liuwei Dihuang Wan had a significant protection effect on type 2 diabetes rats model,and different prescriptions may have different effects and mechanism on the same disease.
