造血干细胞功能异质性研究的“金标准”--单细胞移植技术的建立和优化 被引量:2

摘要 造血干细胞(HSC)是发现最早、在临床中广泛应用并一直发挥范式作用的一类重要的成体组织干细胞.HSC是一个由不同功能亚群组成的高度异质性群体,其自我更新和多向分化潜能主要通过HSC移植这一"金标准"的实验流程来评估和判断.细胞群体水平的研究无法深入探讨其生物学功能,从而使HSC单细胞水平的研究成为必需.为了更好地研究HSC的异质性,本文将详细介绍HSC单细胞移植实验流程及结果检测和分析,旨在促进相关科研人员对于单细胞移植适用性的了解以及具体实验操作的掌握,客观评判具有高度异质性干细胞群体功能,为其他具有高度异质性的干细胞群体的功能研究和评价提供例举和参考. Hematopoietic stem cells(HSC) are one of the earliest, and most extensively studied stem cells that have been used in clinical medicine and serve as an excellent model in stem cell study. HSC are a heterogeneous population consisting of functionally different subsets. HSC transplantation is a golden standard for detection of self-renewal and multi-potential differentiation ability. Nowadays,population-based research cannot meet the need for deep understanding of biological functions and it is necessary to study at the single cell level. In this article, we introduce experimental procedures and data analysis of single cell transplantation. Researchers in related fields may find these technologies useful in their studies. As such, single cell biology is expected to increasingly expand for understanding of fundamental basis in medical science, understand applicability of single cell transplantation, acquire specific experimental operation, and promote wide applications, thus acting as a paradigm and reference for the study of other heterogeneous stem cell population.
