摘要
以MS基本培养基为对照,在培养基MS+6-BA 1.0mg/L和MS+6-BA 1.0mg/L+NAA 0.1mg/L的培养基上接入的山丹小鳞茎无菌苗,应用酶联免疫吸附分析法(ELISA),研究山丹继代培养过程中4种内源激素的变化。结果表明:在继代培养过程中添加的外源激素不同,小鳞茎表现的形态变化也不相同,相对应的是内源激素的含量也发生一定的变化。在MS培养基和同时添加6-BA 1.0mg/L和NAA 0.1mg/L的培养基上小鳞茎ABA含量变化均较为平缓,在添加6-BA 1.0mg/L的培养基上ABA的含量呈现先上升后又呈逐渐下降的趋势;在添加了6-BA 1.0 mg/L和6-BA 1.0 mg/L+NAA 0.1 mg/L外源激素的培养基上小鳞茎的ZR、IAA和GA3含量的高低均与其增殖率呈负相关,而ZR/IAA的比值却与不同培养基中小鳞茎的增殖率呈正相关。当ZR/IAA高时诱导芽的分化,比值低时诱导根的产生。而IAA/ABA比值高时有利于根的分化,比值低时有利于小鳞茎的分化,与ZR/IAA的比值呈负相关。
The small bulbs of aseptic seedlings of Lilium pumilum DC were used as materials after treated with MS,MS+6-BA 1.0 mg/L and MS+6-BA 1.0 mg/L + NAA 0.1 mg/L and MS as a control medium.Changes of four endogenous hormones in Lilium pumilum DC in subculture process were studied by means of ELISA.The results showed that the morphological changes of small bulbs were different in the subculture,and the contents of endogenous hormones were also changed.In the medium of added 6-BA1.0 mg/L+NAA 0.1 mg/L,the change of contents of ABA in the bulbs were relatively flat;the contents in the medium added6-BA 1.0 mg/L ABA first increased and then decreased gradually.In the medium of adding 6-BA 1.0 mg/L and 6-BA 1.0 mg/L+NAA 0.1 mg/L,the contents of ZR IAA and GA3 in the bulbs were negatively correlated with the proliferation rate,but the ratio of ZR/IAA with different medium bulblet proliferation rate were positively correlated.When ZR/IAA was high,the differentiation of buds was induced and the root was induced when the ratio was low.The high IAA/ABA ratio was beneficial to the differentiation of roots.When the ratio was low,itwas favorable to the differentiation of small bulbs and the ratio of ZR/IAA was negatively correlated with the IAA/ABA ratio.
