液相色谱法测定决明子中大黄酚的含量

Content Analysis of Chrysophanol in Cassia Obtusifolia L.by HPLC

建立决明子中降压降脂、保护肝脏的有效成分结合大黄酚的HPLC定量测定方法。方法:选用SUPELCOSI-LTMLC18Column(5μm,4.6mm×250mm)色谱柱,流速1.0mL/min,测定波长254nm,柱温40℃。以大黄酚的含量为测定指标,流动相选甲醇-5%醋酸水溶液(75∶25)。结果:进样量在(0.08～3.20)μg范围内线性关系良好(γ=0.9997)。加样回收率:游离大黄酚为97.45%,RSD为2.21%;总大黄酚为102.14%,RSD为1.85%。结论:本法专属性强,可用于决明子中大黄酚的含量测定。

Objective:To find a quantitative determination method by HPLC for examining the effective content of combining chrysophanol in Cassia obtusifolia L.,which could decrease blood pressure,blood lipid and protect the liver.Methods :SUPELCOSIL TM LC 18 Column (5μm,4.6mm×250mm)was used with flow rate1.0mL/min,detected wave254nm,chromatogratic column temperature40℃and mobile phase MeOH-5%AxOH(75∶25).The content of chrysophanol was detected.Results:The linear relationship was fine(γ=0.9997)in(0.08～3.20)μg.The recovery rate of free chrysophanol was97.45%,RSD2.21%;of total chrysophanol was102.14%,RSD1.85%.Conclusion:This method is specific and can be used to detect the content of chrysophanol.
