摘要
以氢过氧化亚油酸作为脂质过氧化物,亚硒酸钠作为抗氧剂,作用于培养的人脐带静脉内皮细胞,探讨脂质过氧化与抗氧化作用对细胞结构和功能的影响。结果表明,氢过氧化亚油酸能引起细胞收缩、生物膜系统损伤及前列环素合成减少;硒可使细胞脂质过氧化损伤程度减轻。提示:硒可能通过阻止内皮细胞脂质过氧化反应,起抗动脉粥样硬化病变发生的作用。
Linoleic acid hydroperoxide was used to treat cultured endothelial cells(ECs) and selenium as antioxidant, to observe the effect of lipid peroxicationand antioxidation of selenium on the structure and function of ECs. In eachgroup, the concentration of 6-keto-PGF_1α in the medium was determined ECswere observed under phase contrast microscope and EM. The results showed thatunder the increasing concentration of linoleic acid hydroperoxide, the contrac-tion of ECs, dilatation of mitochondria and endoplasmic reticulum, plasmamembrane blebbing were observed. The amount of 6-keto-PGF_1α produced by ECswas significantly reduced. While selenium added can obviously reduce the degreeof ECs contraction, injuries of biological membrane system and amount of PGI_2.
