摘要
为观察铅镉联合作用对大鼠肾小管上皮细胞脂质过氧化的影响 ,对原代大鼠肾小管上皮细胞提取、纯化、鉴定、培养后 ,采用 3× 3析因设计方法进行染毒实验 ,A组 :醋酸铅 0 0 2mmol L ;B组 :醋酸铅 0 1mmol L ;C组 :氯化镉 0 0 0 1mmol L ;D组 :氯化镉 0 0 0 4mmol L ;E组 :醋酸铅 0 0 2mmol L +氯化镉 0 0 0 1mmol L ;F组 :醋酸铅 0 0 2mmol L +氯化镉 0 0 0 4mmol L ;G组 :醋酸铅 0 1mmol L +氯化镉 0 0 0 1mmol L ;H组 :醋酸铅0 1mmol L +氯化镉 0 0 0 4mmol L。染毒时间为 6小时。经超声波粉碎细胞后 ,测定细胞内谷胱甘肽 (GSH)和丙二醛 (MDA)的含量 ,以及谷胱甘肽过氧化物酶 (GSH Px)和超氧化物歧化酶 (SOD)的活性。与对照组比较 ,联合染毒组GSH、GSH Px、SOD水平显著降低 ,MDA水平显著升高 ,析因分析表明 ,铅镉联合作用在对大鼠肾小管上皮细胞GSH、SOD、MDA改变方面有交互作用 (P <0 0 5 ) ,即铅镉联合作用呈现增毒作用 ;在对GSH
In order to observe the combined effects of lead and cadmium on lipid peroxidation,the cultured renal tubular epithelial cells (TEC) of rats are used to perform 3×3 factorial experimental design. In the experimental group (A-H), rats are given lead and cadmium respectively for 6 hours at different concentrations (A: lead acetate 0.02mmol/L; B: lead acetate 0.1mmol/L; C: cadmium chloride 0.001mmol/L; D: cadmium chloride 0.004mmol/L; E: lead acetate 0.02mmol/L+cadmium chloride 0.001mmol/L; F: lead acetate 0.02mmol/L+cadmium chloride 0.004mmol/L; G: lead acetate 0.1mmol/L+cadmium chloride 0.001mmol/L; H: lead acetate 0.1mmol/L+ cadmium chloride 0.004mmol/L). After TEC are smashed by supersonic,the concentrations of glutathione (GSH) and malondiadehyde (MDA) in cells are analyzed. The same analysis is also done for the determination of the activity of glutathione peroxidase(GSH-Px) and superoxide dismutase(SOD). In comparison with control group,significant decrease in activities of GSH、GSH-Px、SOD in cells treated with lead and cadmium and increases of MDA are observed. The data from ANOVA analysis show that the interaction between lead and cadmium on GSH, MDA, and SOD in TEC (P<0.05) is demonstrated while no interaction on GSH-Px is shown.
