大孔树脂纯化“双红”花青素研究 被引量:4

Study on Purification Process of the Anthocyanins from ‘Shuanghong' Vitis amurensis Rupr. by Macroporous Resin

摘要 以"双红"山葡萄为原料,研究NKA-Ⅱ,NKA-9,AB-8,D-101和HPD-100 5种大孔树脂对"双红"花青素的纯化作用。通过静态-吸附解吸试验,筛选出D101树脂为最佳纯化材料,确定最佳吸附-解吸温度为25℃。D101树脂最佳动态吸附-解吸工艺参数为:25℃,上样浓度22.5 mg/m L,上样量8 BV,上样流速为2 m L/min,洗脱剂为75%乙醇-0.01%盐酸,洗脱流速2 m L/min,洗脱剂用量7 BV。该条件下,"双红"花青素纯度为27.78%±0.043%。 Using ‘Shuanghong' Vitis amurensis as raw material, five different macroporous resins(NKA-Ⅱ, NKA-9, AB-8,D-101 and HPD-100) were evaluated for their use to purify anthocyanins from ‘Shuanghong'. Macroporous resin D101 was selected as the most suitable one by using static adsorption and desorption experiments and its optimum adsorption desorption temperature was 25 ℃. The best dynamic adsorption and desorption parameters using D101 macroreticular resin were as follows: the temperature of 25 ℃, anthocyanins concentration of 22.5 mg/m L, sample loading volume of 8 BV(resin bed volume), the adsorption speed of 2.0 m L/min, 75% ethanol-0.01% hydrochloric acid as optimal concentration of eluent, elution flow rate of 2 mL/min, the desorption solution of 7 BV. Under this condition, the purity of anthocyanins from ‘Shuanghong' was 27.78%±0.043%.
